Fluid andmacromolecule secretion by submucosal glands are believed toplay an important role in antimicrobial defense and surfaceliquid homeostasis. The abnormal secretion of the glands willresult in the turbulence of the physiological function in nasalcavity. Allergic rhinitis is an inflammatory disease which happensin the nasal mucosa and it is characterized by excessivesecretion of glands. The pathogenesis of the allergicrhinitis involves many tache of nerve and immunity andfinally result in high reactivity and high secretion. Highsecretion is also excessive fluid secretion and there area lot of explanations,such as,that all kinds ofcytokines,inflammatory media and neurotransmitters whichact directly or indirectly on the endocrine cells of thenasal mucosa,that participation of the matter of Psubstance and that abnormal metabolism of intracellularcalcium. Obviously,there is more complex foundation ofpathology about the high secretion of nasal mucosa in theallergic rhinitis. Aquaporins (AQPs) are a family oftransmembrane proteins that selectively allow the passageof water through the plasma membrane and they are veryimportant to liquid transport and secretion of some glandsin human body. Up till now, at least eleven aquporins areidentified and they are AQP0~AQP10. AQP5 has closerelations with the secretion of submucosal glands. Objedtive To confirm the expression and distributionof Aquaporin 5 (AQP5) in rat nasal mucosa of experimentalallergic rhinitis and to investigate the relationshipbetween AQP5 and the allergic rhinitis. MethodsTwenty-four healthy Wistar rats,including male or female,weighting 200g-300g were divided into two groups randomly,one group was experimental group (n=12), the other wascontrol group(n=12).Generally sensitized rats inexperimental group were given repeated local boostersensitization into nasal cavity.Twelve normal rat nasalmucosa and twelve nasal mucosa from rat with allergicrhinitis were used.The distribution of AQP5 in normal ratnasal mucosa and nasal mucosa with allergic rhinitis wereobserved by immunofluorescence technique. Furthermore,the expression of AQP5 in normal rat nasal mucosa and nasalmucosa from rat with allergic rhinitis were studied byimmunohistochemical staining.Results ①Hematoxylin and eosin staining showed thatthere was obvious inflammation reaction,a large quantityof glands hyperplasia and acidophils soaked in nasalmucosa from rat with allergic rhinitis. ②Bothimmunofluorescence technique and the immunohistochemicalstaining showed that the distribution of AQP5 in normalrat nasal mucosa was in accordance with that in nasalmucosa from rat with allergic rhinitis on the whole.AQP5expressed mainly in the membrane and cytoplasm of theepithelium in the glands,ducts and cilia. ③Thestatistical analysis of the immunohistochemical stainingshowed that the quantity of AQP5 in the rat nasal mucosawith allergic rhinitis(156.37 ±1.93) was obviouslyhigher than that in the normal rat nasal mucosa(178.52±1.94).There was statistical significance between twogroups(t=28.08,p﹤0.01).
