| Objective:Through clinical curative effect evaluation and animal experimental study of using ultrasonic atomization inhalation atomization agent rhinitis Ⅱ to treat adults-severe persistent allergic rhinitis are studied systematically,and discuss the curative effect of Th2 related regulation mechanism.Methods:Clinical research:(1)would be in 40 patients with inclusion criteria,were randomly divided into treatment group(ultrasonic atomization rhinitis Ⅱ atomizing agent)and the control group(ultrasonic atomization dexamethasone sodium phosphate injection).The changes of the symptoms and signs of AR patients before and at the end of treatment,the changes of the symptoms during the follow-up 1 month after the end of treatment,and the changes of the quality of life scale score of nasal conjunctivitis before and at the end of treatment were used to observe the adverse reactions after treatment.To evaluate the clinical efficacy and safety,the possible mechanisms related to interleukin 4 and total immunoglobulin E in Th2 were discussed.(2)animal experimental study:30 SD rats were selected and randomly divided into 5 groups on average:blank control group,model control group,western medicine control group,Chinese medicine high-dose treatment group and Chinese medicine low-dose treatment group.Ovalbumin was used as sensitizing agent and aluminum hydroxide as adjuvant.After successful modeling,corresponding treatment schemes were given.At the end of treatment,the expression level of interleukin-13 related to Th2 immune regulation was measured by elisa assay,and pathological sections of nasal mucosa were made to observe the pathological changes of nasal mucosa in SD rats after treatment.Results:(1)In the clinical study,no.with rhinitis Ⅱ atomizing agent in the treatment of adult-severe persistent AR,the nasal symptoms(sneezing,runny nose,stuffy nose,nasal itching),signs,and total score were improved(P<0.05),and ease the degree is superior to western medicine dexamethasone group(P<0.05).(2)Chinese medicine rhinitis Ⅱ atomizing agent groups before treatment,at the end of treatment,treatment,the symptoms of over 1 month total score from 10.25 + /-1.94 to 2.00 + /-2.00,three time periods are significantly different(P1,P2,P3<0.05),there was a significant difference compared with the western medicine group(P<0.05).(3)The RQLQ scores and total scores of the Chinese medicine group before and after treatment were statistically significant(P<0.05),which was significantly different from that of the western medicine group(P<0.05).(4)The total effective rates of the traditional Chinese medicine group and the western medicine group were 95% and 80%,respectively.(5)There was no significant difference in the level of total IgE between the Chinese medicine group and the western medicine group(P>0.05),but it was statistically significant compared with the control group(P<0.05).(6)Il-4 concentration level in the traditional Chinese medicine group was different from that in the control group and the western medicine group(P<0.05),while there was no difference between the western medicine group and the control group(P>0.05).In the AR model animal experiment of SD rats,(7)There were significant differences in nasal symptoms between different groups before and after treatment(P<0.01).There was no significant difference between the two groups(P>0.05).(8)After treatment,the overall difference of Il-13 concentration in each group was statistically significant(P<0.01).The concentration in the model group was higher than that in other groups,and the difference was statistically significant(P<0.05),indicating that the modeling was successful.Il-13 could be reduced in high-dose group,low-dose group and dimeter group.There was no significant difference among the groups of high-dose TCM,low-dose TCM and dimeter(P>0.05).Conclusion(s):(1)On the clinical application,rhinitis Ⅱ,atomizing agent for the treatment of lung spleen-deficiency type adults-severe persistent AR has significant curative effect,significant relief of nasal symptoms,improve patient quality of life,no adverse reactions and side effects,low recurrence rate and 1 months.(2)In the clinical study,rhinitis Ⅱ atomization agent number AR to treat it that can reduce the total IgE,IL-4,thus inhibiting Th2,reach the role of immune regulation.AR(3)In SD rats models in animal experiments,rhinitis Ⅱ atomization agent can reduce IL-13,number associated with the immune regulation of Th2,thus inhibiting allergic inflammation.(4)The action mechanism of rhinitis II atomizer may be related to the Il-4 / Il-13 /Jaks/STAT6 signaling pathway,which may act on the Il-4 receptor and play an antagonistic role.Therefore,it can down-regulate the expression of total IgE,Il-4 and Il-13,thereby inhibiting the over-differentiation of Th2 and alleviating the allergic inflammatory reaction. |
