| Background: Oral Lichen Planus(OLP)is a chronic inflammatory disease that occurs in the oral mucosa,including erosive and non-erosive types.The WHO defines it as a precancerous state.Patients with erosive OLP are often accompanied by mucosal pain,which affects eating and speaking,and has certain harm to the patient’s physical and mental health.The etiology and pathogenesis of OLP have not yet been fully elucidated.The role of genetic factors in the pathogenesis of OLP cannot be ignored,studies have reported that the polymorphism of certain genes may be related to the susceptibility of OLP.Interleukin-23 receptor(IL-23R)and interleukin 33(IL-33)play an important role in the pathogenesis of various chronic inflammation and immune diseases.Interleukin23(IL-23)works by binding to IL-23 R,and after binding,it activates downstream signaling pathways and exerts biological functions through its N-terminal immunoglobulin domain.IL-33 is a polymorphic cytokine that has a dual role in the body.First,it can act as a cytokine of the interleukin-1(IL-1)family outside the cell,and induce pro-inflammatory immune response and cytokine activation by binding to the orphan receptor ST2,a member of the IL-1 receptor family.produce;secondly,it acts as a cell’s nuclear factor.After the cell is injured,stressed or cell death,IL-33 will be released from the nucleus,thus acting as an "alarm".Object: This study aims to explore the relationship between IL-23 R gene and IL-33 gene polymorphisms and genetic susceptibility to OLP in the Han population in Qingdao,Shandong,so as to provide a new theoretical basis for the etiology of OLP.Method: Using the case-control research method,223 subjects were selected and 106 were in the case group,including 31 in the erosive type and 75 in the non-erosive type;117 in the healthy control group.Collect 2 m L of whole saliva from the study subjects,and use Taq Man Real-time Quantitative PCR technology to amplify the DNA in the exfoliated epithelial cells in the whole saliva,and read the results directly through SDS v2.3 software.Use SPSS 22.0 statistical software to analyze and compare the differences in allele frequency and genotype distribution frequency between IL-23 R gene rs7517847 locus and IL-33 gene rs3939286 locus,and based on different inheritance modes(Co-dominant mode,Dominant mode,Recessive mode,Over-dominant mode,Log-additive mode)to perform genotype association analysis between OLP and IL-23 R rs7517847 locus and IL-33 rs3939286 locus.Result:1.Three genotypes of TT,GT and GG were detected at the IL-23 R gene rs7517847 site;two genotypes of CC and CT were detected at the IL-33 gene rs3939286 site.2.The genotype distribution of IL-23 R rs7517847 and IL-33 rs3939286 loci in the case group and the control group were in accordance with the Hardy-Weinberg(H-W)equilibrium law(P>0.05).3.IL-23 R rs7517847,IL-33 rs3939286 locus allele frequency and genotype distribution frequency were not statistically different between case group and control group,erosion group and control group,non-erosion group and control group(P>0.05).4.For IL-23 R rs7517847 locus in the over-dominant genetic mode(GT vs GG+TT),the difference in genotype distribution frequency between the non-erosive group and the control group was statistically significant(P<0.05),the risk of individuals with GT genotype is 1.81 times that of individuals with TT+GG genotype(OR=1.82,95%CI: 1.00-3.29).Conclusion:1.There are genetic polymorphisms in IL-23 R rs7517847 and IL-33rs3939286 in the tested population.2.IL-23 R rs7517847 gene polymorphism may be related to the genetic susceptibility and severity of OLP disease,and GT genotype carriers may have a higher risk of disease.3.IL-33 rs3939286 gene polymorphism has no obvious correlation with OLP disease genetic susceptibility. |
