Leptin As A Survival Enchance Factor In Grainy Fat Grafting

Grainy fat grafting is a useful method in body sculpture. In such operation, the major concern is the survival rate of the grafted fat. Many scholars are striving for the solution.Lepin is one of the factors secreted by fat cell. It can enhance tissue angoigenesis besides regulating the body energy balance. In this study, we tested the effects of leptin on the survival of grainy fat grafts in animals and inspected other correlative problems with clinic and animal experiments.Wistar rats were used in the experiment, which comprised 4 parts. In part one, we treated grainy fat with leptin, saline and MEM before subcutaneous grafting. After 10 days, the weight-maintence of the grafted tissue was 108.3% in the group which 0.6g tissue was processed with 20ng/ml leptin. Compared with other processing methods, this created an obviously high weight-maintence rate. In part two, we observed angiogenesis in the grafted tissue with AKP, Actin and VEGF staining. It was found that leptin could enhance the angiogenesis of the grafted tissue. In part three, we compared the rat serum leptin level before and after operation with ELISA. The results testified no obvious difference in serum level between these two times. The last part of the experiment tested the influence of the subcutaneously administered leptin on the serum leptin level. After processing the grainy fat with 2ml leptin (20ng/ml), we grafted the fat subcuteneously. 10 days after the operation, the serum leptin level was little higher than that before operation without statistic difference.14 patients were solicited to attend the clinical experiment. The patients' blood samples were obtained before and after liposuction. The aspirated liquid in liposuction was harvested. The patients' body mass index (kg/m~2), subcutaneously injected liquid before liposuction and the aspirated fat were recorded.
