A Study On The Effective Of Leptin On The Survival Rate In Grainy Fat Transplantation With Sd Rat

Objective This experiment is to study the effect of the different densities leptin on the survival rate of grainy fat transplantation.Methods Different densities leptin was used as interference in grainy fat transplantation on experimental animal models. The transplanted tissue was separate taken out after 7 , 14 , 28 and 56 days. Being weighted and calculating the remained-weight rate, the tissue was dyed with HE staining and CD34 staining and VEGF staining. Microvessels densities both on experimental group and controlled groups were quantitatively researched in various periods. The growth course of vessels was observed. The data was analyzed with SPSS 17.0 software.Results 1. The fibrous membrane around the tissue in leptin group was thin and there was less fat cell necrosis , compared with the controlled group. The remained-weight rate of NS group,10ng/ml leptin group, 30ng/ml leptin group and 50ng/ml leptin group at 7 days after the surgery was 82.5%,102.5%, 121.5%,133.5%;after 14 days was 69.5%,83.5%,91%,93.5%, after 28 days was 47%,70.5%,80.5%,83%, after 56 days was 40.5%,61%,69.5%,72.5%. The differences were statistic significant between the leptin groups and the control ( P < 0105) . The examinations of the HE and VEGF stain showed that there was the rich vascularization in leptin treated tissue.2. Microvessels can be observed clearly. The values of microvessel density in 50ng/ml leptin groups were 72±3.24,62.4±6.02,58±3.08,47±5.52/mm2 at 7 , 14 ,28and 56 days after the surgery. This was obviously high as compared with saline groups and the other control groups in the same observing periods ( P < 0.05) . CD34 staining in leptin groups was also intense.Conclusions 1. There are significant differences between the NS group and the leptin groups in7, 14, 28 and 56 days. And with the higher density of the leptin, the more effective on promoting vascular of tissue hyperplasia, the higher remained-weight rate. Among it, in the 50ng/ml leptin group, both the vascular density and the remained-weight rate are better than the others, and the higher survival rate of the transplanted tissue.2. It is decline that the vascular density and the remained-weight rate following the time. During the experiment, It is the very time of 7 days that there is the most powerful of new tissue-vascular and the highest remained-weight rate. And also, the studies have shown that 7 days is the critical period for measurement on the survival rate of transplanted tissue.
