Objective To study the effect of Electrochemical Therapy(EChT) with low milli- Ampere(lOmA) on apoptosis and cell cycle of liver cancer cell lineSMMC7721.Materials and Methods Cultured liver cancer cells SMMC7721 in vitro were treated with EChT. Equal number of cells was put into 24-well plate. They were divided into control group and treated group in random. Treated group was subdivided into 4 groups according to the different charge: 3C, 5C, IOC, 20C(C: coulomb), 3 wells per group. During experiment, two parallel platinum electrodes were inserted into the liquid with cultured cell, 5 mm depth. The distance between two electrodes was 1.0 cm. Control group did not treated with EChT. 6 hours and 24 hours after treatment, the rate of apoptosis and the percentage of cell cycle were detected by flow cytometry. The average data of each 3 wells were used. Results Six hours after treatment, the rate of apoptosis in control group was 0.05%, and in treated group with different charge (3C, 5C, IOC, 20C)were 0.56%, 0.87%, 1.09%, 1.63% respectively. The rate of apoptosis in treated groups did not increase obviously, but compared with control group, still had statistical change (P<0.05). The percentage in different cell cycle phases showed no significant change (.P>0.05). 24 hours after treatment, the rate of apoptosis in control group was 2.91%, and in treated groups with different coulomb (3C, 5C, IOC, 20C) were 12.59%, 22.19%, 46.16%, 56.68% respectively. The rate of apoptosis in treated group increased significantly (PO.05). The percentage of G0-G, phase in 20C treated group increased obviously (74.03%). The percentage of GQ-G{ phase in treated group had an increasing tendency as coulomb increased, and the percentage of S phase had a decreasing tendency. Conclusions Within six hours of low mA EChT, The rate of apoptosis in treated groups did not increase obviously, but 24 hours after EChT, showed significant change between control group and treated group(P<0.05). Within 6 hours, the percentage of cell cycle did not changed obviously, but 24 hours after EChT, the percentage of G0-G, phase had an increasing tendency, and the percentage of S phase had a decreasing tendency .The highest percentage of GO-G! phase was in 20C treated group (74.03%).
