Effect Of Yiqi Warming Yang Formula On Foxp3 ~ + Treg - Immune Tolerance Pathway In Allergic Rhinitis

Objective To explore Yiqi Wenyang Decoction regulate the amount of Treg cell and the immune tolerance of allergic rhinitis, to achieve the purpose of treatment.Methods In this study 48 SD rats as the experimental animal, divided into blank group, model group, high, middle and small dose of Yiqi Wenyang Decoction group, and loratadine group,8 rats in each group. To establish the model of allergic rhinitis in rats, using immunohistochemistry, flow cytometry, Western Blot, PCR and other technology observed Yiqiwenyang Decoction on Foxp3+mRNA and protein expression of Foxp3+, Treg and cytokines influence.Results:1 symptoms:Yiqi Wenyang Decoction and loratadine significantly on allergic rhinitis in rats after administration of 0 to 14 days (compared with the model group, the curative effect of P< 0.05), but in 14-28 days after administration of Yiqi Wenyang Decoction effect is better than loratadine effect (P>0.05); 2 the pathological aspects:Yiqi Wenyang Decocrion had a significant improvement on rats with allergic rhinitis in sneeze, scratching and other symptoms and nasal inflammation status. The high and middle dose of the drug had the best effect (compared with model group, P<0.0001); 3 cytokines:Yiqi Wenyang Decoction on IL-10, TGF-beta 1 had significant promoting effect of increasing and the inhibition of IL-4, IL-13, but in IL-4 and the control group of loratadine is equivalent (P>0.05).4 Treg cells flow cytometry results:CD4+CD25+Foxp3+Treg cell values decreased significantly in allergic rhinitis rats, in Yiqi Wenyang Decoction and loratadine treatment significantly restored near normal (compared with the blank group, P>0.05); 5 Western Blot testing results:generation of Foxp3 transcription factor allergic rhinitis mouse decreased obviously, in the treatment of Yiqi Wenyang Decoction the Foxp3 transcription factor increases, and the effect is better than loratadine; 6 results of RT-PCR:the expression of Foxp3+mRNA in allergic rhinitis rats decreased significantly, in the treatment of Yiqi Wenyang Decoction induced Foxp3+mRNA expression increased significantly, and the effect is better than loratadine.Conclusion:Yiqi Wenyang Decoction can significantly control the symptoms of rats with allergic rhinitis, improve the nasal mucosa epithelium infiltrating eosinophils, up-regulated Foxp3+mRNA and Foxp3 transcription factors, the number of Foxp3+CD4+CD25+Treg cells induced by increased, thus IL-10 and TGF- beta 1 increased, the inhibition of Th2 cell activity, reduce the generation of IL-4 and IL-13, and then inhibit the immune inflammatory reaction followed by a series of symptoms, reducing the treatment of disease.The research significance from a new angle of treg-immune tolerance to know of Yiqi Wenyang Decoction in the treatment of allergic rhinitis mechanisms, namely by improving immune tolerance ability, improve the body of highly sensitive state, so as to alleviate the symptoms. Provides a new experimental basis for clinical application of Yiqi Wenyang Decoction.
