The Effect Of Yiqi Wenyang Decoction On Chemotaxis And Differentiation Of NK Cells In Allergic Rhinitis Mice In Vitro

Objective:To observe the effect of Yiqi Wenyang Decoction on the chemotaxis and activation of NK cells in allergic rhinitis mice in vitro,and to explore the possible mechanism of Yiqi Wenyang Decoction in the treatment of allergic rhinitis.Methods:30 mice were randomly divided into blank group,model group,low,middle and high dose group of Yiqi Wenyang formula and cetirizine group,with 5 mice in each group.After AR model was established,normal saline was given to blank group and model group,water decoction of low,middle and high concentration of Yiqi Wenyang Decoction was given to traditional Chinese medicine group,cetirizine was given to cetirizine group,and water solution was given to cetirizine group Preparation of blank,model,low,medium and high dose of traditional Chinese medicine,cetirizine containing serum;taking normal mice,killing and separating spleen,separating and purifying NK cells;using dust mite as external stimulant,using Transwell method and CCK-8 method to detect the migration ability of NK cells,using ELISA method to detect the expression of IL-5,IL-10,IL-4,IFN-y in NK cells.One way ANOVA(one way ANOVA)analysis method was used to compare the differences among groups,and Bonferroni method was used to compare the two groups with the same variance.Results:1.Chemotaxis of NK cells in Allergic Rhinitis Mice:the number of NK cells in the upper chamber(Transwell chamber)decreased significantly after adding stimulants to the lower chamber.The number of NK cells in the upper chamber(Transwell chamber)increased significantly after adding stimulants to the lower,middle and high dose groups containing Yiqi Wenyang decoction,but the low and high dose groups of Yiqi Wenyang Decoction did not return to normal The level of blank group,but there was no significant difference between the middle dose group of Yiqi Wenyang Decoction and blank group(P>0.05).The effect of middle dose group of traditional Chinese medicine on inhibiting the chemotaxis of NK cells to the lower chamber added with stimulants was significantly higher than that of high dose group,and the effect of traditional Chinese medicine on the chemotaxis of NK cells in the low dose group was similar to that in the middle and high dose groups(P>0.05).The effect of Chinese medicine group was the same as that of cetirizine(P>0.05).2.Differentiation of NK cells in mice with allergic rhinitis:After external stimulation,the concentrations of IL-4,IL-5 and IFN-? of NK2 type cytokines increased significantly,and the concentrations of IL-10 of NK1 type cytokines decreased significantly.Yiqi Wenyang Decoction inhibited the secretion of IL-4 and IL-5 in low,medium and high doses,and promoted the secretion of IL-10 in NK1,but the concentration of IL-4,IL-5 and IL-10 did not return to the level of normal blank group.The effect of inhibition of IL-5 and IL-4 in middle dose group of traditional Chinese medicine was better than that in other two groups,The effect of middle dose group of traditional Chinese medicine on the release of IL-5 and IL-4 was similar to that of cetirizine(P>0.05).On IL-10,the serum IL-10 concentration in the middle dose group of traditional Chinese medicine was significantly higher than that in the other two groups,with the best effect,while cetirizine had the same effect on IL-10 secretion of NK? cells as that in the middle dose group of traditional Chinese medicine;on IFN-y,each dose group of traditional Chinese medicine had no effect on IL-10 secretion as that of cetirizine.Conclusion:Yiqi Wenyang decoction can obviously inhibit the chemotaxis of NK cells to inflammatory sites in mice with allergic rhinitis,reduce the migration of NK cells to the pathological site and release NK2 type inflammatory cytokines to aggravate the inflammatory response,and can down regulate the IL-5,IL-4 and other inflammatory cytokines released by NK2 type cells in mice with allergic rhinitis,and promote the secretion of NK1 type cytokines IL-10,so as to regulate the biased state of NK2 type reaction,and then inhibit a series of subsequent changes State reaction.It is suggested that Yiqi Wenyang decoction can treat allergic rhinitis by regulating the chemotaxis of NK cells and the release of cytokines.This experiment also shows that cetirizine can also be used to treat allergic rhinitis through this mechanism.
