Objective: By observing the TD model rats weight,changes in behavior and brain tissue of Synapsin Ⅰ,the positive expression of PSD-95,confirmed that Jianpi Rougan Xifeng decoction on the role of synaptic plasticity,Jianpi Rougan Xifeng decoction on the mechanism of action of TD model rat neural synaptic transmission.Methods: 60 SD rats were randomly divided into blank control group,model control group,haloperidol group,Jianpi Ruogan Xifeng decoction low dose group,Jianpi Ruogan Xifeng decoction middle dose group,Jianpi Ruogan Xifeng decoction high dose group.The model was established by intraabdominal injection of IDPN for 7 days.After 6 weeks of intragastric administration,the body weight of the rats was measured at the 0 week,2week,4 week and 6 week after administration,and the average body weight of the rats was obtained.The positive expression of synaptic associated protein Synapsin Ⅰ and PSD ≤ 95 was detected by immunohistochemistry.Results: 1.The body weight of rats was measured at 0 week,2 week,4week and 6 week after administration,and the average body weight of rats was obtained.the results showed that the body weight of rats in blank group was significantly higher than that in other groups(P < 0.05).2.HE staining showed that after 4 weeks of administration,the granulosa cells and glial nucleus atrophy and softened foci could be seen in the model group after 4 weeks of administration,western medicine group,low dose group of traditional Chinese medicine,middle dose group of traditional Chinese medicine,low dose group of traditional Chinese medicine,middle dose group of traditional Chinese medicine,low dose group of traditional Chinese medicine,middle dose group of traditional Chinese medicine.There was no significant difference between the high dose group and the normal group.3.Under 400 fold microscope,the expression of PSD-95 in the sections of the module group was significantly lower than that of the others.The expression of PSD-95 in the other five groups was the strongest,the high dose group was the second,the middle dose group was the second,and the western medicine group and the low dose group were the weakest,and there was no significant difference between the two groups.Under 400 fold microscope,there was no significant difference in the expression of Synapsin Ⅰ in model group,blank group,western medicine group,low dose group,middle dose group and high dose group.Conclusion: IDPN model can be used as an animal model of tic disorder to a certain extent.IDPN modeling treatment is the cause of weight gain in rats,while haloperidol and Jianpi Ruogan Xifeng decoction have no effect on body weight gain in TD model rats.Jianpi Ruanggan Xifeng decoction and haloperidol have protective and repair effects on striatum of rats.Jianpi Ruogan Xifeng decoction can increase the expression of PSD-95 in striatum of TD model rats.The target of Jianpi Ruogan Xifeng decoction in the treatment of tic disorder rats is that there is no change in Synapsin Ⅰ in striatum of rats after PSD-95.IDPN modeling. |
