Objective:Based on the software "Traditional Chinese Medicine Auxiliary Platform"(V2.5),to explore the prescription and medication rules of Professor Zhao Fuhai for the treatment of Post-percutaneous coronary intervention angina,to explore his treatment concept,and to analyze the professor’s best core drugs for the treatment of this disease through the method of network pharmacology,in order to obtain the main active components,targets and possible signal pathways of the drug for the prevention and treatment of angina pectoris,and to explore its mechanism of action in the treatment of postoperative angina pectoris.Methods:1.From January 2021 to January 2022,Professor Zhao Fuhai’s outpatient diagnosis and treatment of patients with recurrent angina pectoris after PCI were collected,and they were screened in strict accordance with the stringent criteria,and 83 qualified medical records were finally selected.Through the traditional Chinese medicine inheritance platform software,two people and two machines independently input medical record information,established Professor Zhao Fuhai’s treatment of postoperative angina pectoris database,and use the platform’s improved mutual information method,association rules and other functions to analyze the four natures and five flavors,and channel tropisom of the drugs used in the prescription,the frequency of medication,the combination of drug pairs and other statistics,association rules analysis,summed up Professor Zhao Fuhai’s prescription rules and treatment concepts for the treatment of the disease,and obtained the best core drug pairs;2.The potential active ingredients of the core herb pair Rhizoma Chuanxiong-Radix Paeoniae Rubra obtained through the TCMSP database and literature supplementary collection data mining part were imported into the Swiss Target Prediction database for target screening,and the disease-related effects were obtained in the Disgenet and Genecards databases.Then use R language software to screen the intersection of drug and disease targets,draw Venn diagram,and use Cytoscape software to construct"drug-component-target-disease" network diagram and protein-protein interaction network diagram,The core targets were analyzed,and R language software was used to conduct enrichment analysis of biological processes and metabolic pathways.Results:1.A total of 83 medical records were included in this study.The gender statistics showed that the majority of patients with recurrent angina pectoris after PCI were male,mainly in the age group of 55-74 years old.The top 10 symptom frequency statistics are chest tightness,chest pain,fatigue,palpitations,dizziness,shortness of breath,dry mouth and bitter taste,wheezing,and frequent urination respectively.The top 3 signs of tongue statistics are tongue dishes,pale red tongue,and scalloped tongue,and the top 3 frequencies of tongue coating are respectively White and greasy fur,yellow and greasy fur,and thin white fur;the top 3 pulse patterns are stringy,stringy,and slippery,and the top 3 syndrome types are phlegm and blood stasis syndrome,qi deficiency and blood stasis syndrome,respectively.Syndrome with phlegm-heat accumulation;a total of 155 drugs were used,and there were 35 drugs with a cumulative use frequency of>10 times.The top 10 frequencies from high to low are Chuanxiong,Salvia,red peony,safflower,peach kernel,Poria,Gualou,Astragalus,Angelica,and Xiebai;the commonly used drugs are traditional Chinese medicine,with the main medicinal properties being warm,followed by cold;the medicinal flavors are mainly sweet,bitter,and pungent;the medicines are mainly attributed to the liver,heart and spleen meridians,and the best core drug pair is obtained through association rule analysis,for the Rhizoma Chuanxiong-Radix Paeoniae Rubra.2.The top 5 core targets obtained by PPI network are TNF,AKT1,GAPDH,VEGFA,EGFR;GO analysis results show that biological process pathways are mainly enriched in oxidative stress response,inflammatory response regulation,protein serine/threonine Up-regulation of acid kinase activity,peptidyl-tyrosine phosphorylation,peptidyl-tyrosine modification,regulation of MAP kinase activity,epithelial cell proliferation,and cellular response to chemical stress;the enriched cellular components expressed mainly in Membrane rafts,membrane microdomains,membrane domains,nuclear envelope,outside of plasma membrane,collagen-containing extracellular matrix,plasma membrane rafts;the enriched molecular functions are endopeptidase activity,protein tyrosine kinase activity,phosphatase binding,Protein phosphatase binding,hormone receptor binding,serine hydrolase activity,drug binding.The important information pathways obtained by KEGG enrichment analysis are PI3K-Akt signaling pathway,proteoglycan in cancer,MAPK signaling pathway,Rapl signaling pathway,Ras signaling pathway,lipid and atherosclerosis,and AGE in diabetic complications-RAGE signaling pathway,Kaposi’s sarcoma-associated herpesvirus infection and other pathways.Conclusion:1.Professor Zhao Fuhai believes that the main pathogenesis of recurrent angina pectoris after PCI is the deficiency of the standard,the disease is mainly associated with the heart,and it is related to the liver and spleen.The treatment concept can be summarized as follows:(1)cultivating primordial and eliminating pathogens,and treating both the symptoms and the symptoms;(2)The phlegm and blood stasis are eliminated and the blockage is opened;(3)the emotions are adjusted and double heart therapy can be chosen2.Paeonia lactiflora has multiple mechanisms to prevent and treat postoperative angina pectoris through anti-apoptosis,promotion of vascular proliferation,anti-inflammatory and anti-oxidative stress. |
