Objective: Taking traumatic femoral head necrosis model rats as the research o bject,Taohong Siwu decoction was used to intervene in the model group,and the effect of Taohong Siwu decoction on Notch1 receptor in traumatic femoral head necrosis model rats was observed,so as to explain Taohong Siwu decocti on promoting trauma Mechanism of neovascularization and bone regeneration in the rat model of femoral head necrosis.Method: All 60 SPF experimental rats were fed into the experimental group f or 1 week,and then divided into 5 groups according to random methods,nam ely Taohong Siwu Tang high,middle and low three groups,model group and s ham operation control group.12 animals per group.The Taohong Siwu decocti on group and the model group were modeled,and the sham operation group w as incised to expose the hip joint capsule and sutured directly.One rat was ra ndomly selected from each group 4 weeks after the operation.,Color,and arti cular cartilage were roughly observed to determine whether the modeling was s uccessful.The rats in the Taohong Siwu Decoction group were given the same amount of high,medium and low concentration Taohong Siwu Decoction after4 weeks after surgery.The group and sham operation group were given the same amount of normal saline.After 8 weeks of anesthesia,the rats in each group were sacrificed and the femoral head was excised.After fixation,decalcification,dehydration and embedding,the sections were stained and analyzed under light microscope The number of total femoral bone pockets and empty bone pocket s in the femoral head of the rats in the group was calculated,and then the av erage empty bone pocket rate was calculated,and the expression of the averag e optical density of Notch1 was detected by immunohistochemistry for statistica l analysis.2.Results:1.The average empty bone lacunae rate of Taohong Siwu Tang’s hi gh,middle and low groups is significantly lower than the model group,but higher than the sham operation group(P <0.05),the difference is statistically significant;2.The average optical density expression of Notch1 receptor in Taohong Siwu Decoction rats was significantly higher than that in model group and sham ope ration group(P <0.05),the difference was statistically significant;3.The average empty bone lacuna rate of Taohong Siwu Tang low-dose group was higher than Taohong Siwu Tang high-and middle-dose group(P <0.05),the difference was statistically significant;There was no significant difference inthe average empty bone lacuna rate in rats(P> 0.05);4.The average optical density expression of Taohong Siwu Tang low-dose group rats is lower than Taohong Siwu Tang high-and middle-dose groups(P <0.05),the difference is statistically significant;There is no obvious differen ce in optical density expression(P> 0.05);Conclusion:Taohong Siwu Decoction can up-regulate the expression of Notch1 receptor in traumatic femoral head necrosis model rats and reduce the rate of empty bone lacuna to promote new blood vessels and new bone formation;an d the low-dose group up-regulate the expression of Notch1 receptor and reduce empty bone The rate of dimple rate was not as good as the middle and high dose groups,and there was no significant difference between the middle and h igh dose groups.
