| Objective: Through detection of Taohong Siwu Decoction on traumatic avascular necrosis of the femoral head model in peripheral blood EPCs expression effect of Taohong Siwu Decoction in the treatment of traumatic avascular necrosis of the femoral head in the specific mechanism of action, and to provide scientific basis for the Taohong Siwu Decoction in the treatment of traumatic avascular necrosis of the femoral head, but also for the clinical treatment of ANFH by looking for new drug targets.Methods: 29 rats, after 1 weeks of feeding were randomly divided into three groups, 8 rats in normal control group, sham operation group 8, model group 13, model group in accordance with the round ligament broken with the dissection of the model, after 4 weeks, the three groups were randomly selected 3 rats that was gross morphological and histological observation, testing the success of modeling, modeling success after the model group were randomly divided into 2 groups, namely Taohongsiwu Decoction group and model group, 5 rats in each group, Taohongsiwu Decoction group with Taohong Siwu Decoction orally, the remaining three groups were fed with the same volume of normal saline the drug, after 8 weeks of treatment of femoral head gross observation, histological observation, and test the EPCs in the peripheral blood of rats(CD34+, CD133+) level.Results: 1、Femoral head gross morphological observation: compared with sham operation group, control group, Experiment 4 weeks after the module of articular cartilage surface color slightly dark, collapse of the femoral head deformation. Liang Jun;2、Morphological Observation: modeling group bone marrow, bone necrosis, bone trabecular structural abnormalities, empty bone lacunas quantity increase or osteogenic cell nuclear pyknosis, indicating that the rat traumatic femoral head necrosis model has been established. In the model group, Taohong Siwu Decoction group gavage for 8 weeks after the empty bone lacuna rate was significantly higher than that in normal control group(P < 0.05); model group, Taohong Siwu Decoction group gavage for 8 weeks after the empty bone lacuna rate were significantly higher than those in the sham operation group(P < 0.05); intragastric administration 8 weeks of sham operation group, normal control group had no significant difference; model group gavage for 8 weeks of empty bone lacuna rate higher than Taohong Siwu Decoction group(P < 0.05);3、Peripheral blood EPCs(CD34+and CD133+) to detect the number of: model group and normal control group, sham operation group compared to EPCs(CD34+and CD133+) levels decreased significantly, the difference is statistically significant(P < 0.05), compared with the model group, differences in the level of Taohong Siwu Decoction in EPCs group(CD34+and CD133+) have significant difference(P < 0.05).Conclusion: we have successfully established the model of traumatic femoral head necrosis in rats, Taohong Siwu Decoction can obviously promote traumatic femoral head necrosis in a rat model of peripheral blood EPCs(CD34+and CD133+) level of expression, and further from the microscopic and molecular level description of its prevention and treatment for traumatic avascular necrosis of the femoral head. |
