Objective: To explore the mechanism of Taohong Siwu Decoction in delaying the progression of traumatic femoral head necrosis by observing and detecting the pathological morphology and Smad1,Smad5,Smad8 protein and m RNA expression in the model of traumatic avascular necrosis of the femoral head.Method: 30 SD male rats were randomly divided into blank group,modeling group,Taohong Siwu Decoction high-dose,middle-dose and low-dose groups,a total of 5groups,with 6 rats in each group;modeling group and Taohong Siwu Decoction three groups of intervention group rats After 4 weeks of modeling by stripping the blood supply of the femoral head,the rats in the Taohong Siwu Decoction low,middle and high groups were given 4,8,and 16 g/kg Taohong Siwu Decoction respectively,and the blank group and the model group were given gavage.The same amount of normal saline,twice a day for each rat,was administered by gavage for 6 weeks;after 6weeks of intervention,the femoral head bone tissue of the rats was removed,and HE staining was used to observe the trabecular bone,osteocytes,hematopoietic cells and count the rate of empty bone lacunae.The expression of Smad1/5/8 protein in bone tissue was detected by western blot,and the m RNA expression of Smad1/5/8 in cells was measured by RT-PCR.Result:Under the HE staining microscope,the calcified tissue,trabecular bone,and hematopoietic cells of the femoral head of the Taohong Siwu decoction group were improved compared with the model group.Significant(P<0.05);the protein content of Smad1,Smad5 and Smad8 in bone tissue of each group,Taohongsiwutang high,medium and low dose groups were higher than the model group,the data difference was statistically significant(P<0.05);The m RNA expression levels of Smad1,Smad5,Smad8 in tissue cells of each group were higher than those in the high-dose,mediumdose and low-dose Taohongsiwutang groups than in the model group,and the differences were statistically significant(P< 0.05).Conclusion:Taohong Siwu Decoction can increase the relative expression levels of Smad1,Smad 5,Smad 8 protein and m RNA in traumatic femoral head necrosis in SD male rats,and improve the pathological morphology of the femoral head in the rat model,suggesting that the prescription may relieve traumatic injury through Smads signaling pathway.Femoral head necrosis.
