Benign prostatic hyperplasia(BPH)causes prostatic cell and fibrosis because of aging and functional testis in elderly man.It is one of the factors that seriously affects the quality of life of middle-aged and elderly men.It causes urgency,urinary frequency,dysuria and a series of urinary tract symptoms.With the rapidly development of Chinese aged society,more and more middle-aged and old men suffer from this disease.Therefore,timely and effective treatment is of great significance to BPH patients.Acupuncture,moxibustion and other traditional Chinese medicine treatment of BPH have achieved good curative effect,while its mechanism is seldom studied.The role of acupuncture in treating BPH and regulating the level of sex hormone to influence the relationship between prostate cells need to be further explored.BPH is closely related to apoptosis of prostatic cells,and the relationship between BPH and prostatic apoptosis is still to be explored,which provides a scientific basis for acupuncture treatment of BPH.ObjectiveThe main points for the treatment of urinary system diseases in Jin's-3 needle therapy are "three needles of urine"(Guan Yuan,ZhongJi and San Yin jiao).Clinically,it is effective to treat BPH with a combination of syndrome differentiation and acupuncture.The mechanism of acupuncture treatment of BPH is not quite clear.Recent years,studies on BPH indicate that prostatic hyperplasia is closely related to apoptosis of prostate cells,and cell proliferation is not obvious.In this experiment,we studied acupuncture effect for BPH model rat prostate wet weight and index(PI),prostate tissue morphology,cell apoptosis rate of prostate and apoptosis related proteins the Bcl-2 and the influence of the expression of bax to explore the related mechanism of acupuncture treatment of BPH and provide scientific basis for clinical treatment of BPH.MethodIn this experiment,rats were divided into 3 groups by random: normal group,model group and acupuncture group.The BPH model and acupuncture group was established by continuous subcutaneous propionate injection for 28 days to uncastrated rats.The normal group was injected with a corresponding amount of olive oil under the skin at the same time.From the 29 th day,the acupuncture group was engaged in acupuncture intervention,while the model group and the normal group were engaged for 21 consecutive days at the same time.After the last intervention,all rats were fasting for 12 hours and water was forbidden for 6 hours,and the abdominal aorta was taken after weighing.The complete prostate tissue was taken out and weighed to calculate the prostate index(PI).HE staining was used to observe histological morphology.TUNEL method was used to detect apoptosis cells,and apoptosis rate was calculated.Immunohistochemical method was used to detect the expression of apoptosis-related proteins bcl-2 and bax.Results1.The results of the calculation of wet weight and PI in the prostate were: compared with the normal group,the wet weight of prostate in the model group and the acupuncture group was significantly increased(P< 0.05),and the prostate index was significantly increased(P< 0.01).Compared with the model group,the prostatic wet weight and prostate index of acupuncture group were significantly decreased(P< 0.01).2.HE staining results: the prostate epithelium of the normal group was single columnar or cuboid,and the adenoepithelial cells were arranged neatly,and the nucleus was located in the basal part,which was round.The regulation of the glandular cavity is smooth,and the glandular cavity is full of pink acid secretion.No fibrous tissue hyperplasia.The rat prostate epithelial cells in the model group were columnar,clustered,closely aligned,and the nuclei were large,round or elliptic.It can be seen that glandular epithelial cells proliferate into the glandular cavity,and the glandular cavity becomes smaller and even closed.There is no pink-stained acid secretion in the glandular cavity.There are a lot of fibrous tissue hyperplasia.The prostate epithelium of the acupuncture group was columnar,a small number of high columnar cells,and some of the cells were arranged neatly,and the nuclei were round.The hyperplasia of the glandular epithelium formed a small number of protuberances in the glandular cavity,the glandular cavity was slightly reduced,and the glandular cavity was slightly reduced with the acid secretion.There is a small amount of fibrous tissue hyperplasia.3.The apoptosis rate of the acupuncture group was significantly higher than the normal group and the model group(P<0.05).The apoptosis rate of the model group and the normal group was not statistically significant(P BBB 0.05).4.Bcl-2 protein test results: compared with the normal group,the expression of bcl-2 protein in the prostate tissue of the model group was significantly increased(P < 0.01).There was no statistical difference between bcl-2 protein expression and normal group in the prostate tissue of acupuncture group(P BBB 0 0.05).Compared with the model group,the expression of bcl-2 protein in the prostate tissue of acupuncture group was significantly decreased(P< 0.01).5.Bax protein test results: there was no statistical difference between the normal group and the model group in bax protein expression(P BBB 0.05).The expression of bax protein in the prostate tissue of acupuncture group was significantly higher than that in the normal group and the model group(P < 0.01).Conclusion1.The BPH rat model can be established by subcutaneous injection of testosterone propionate.2.Acupuncture can reduce the weight of prostate tissue in BPH rats.3.Acupuncture can improve the change of prostate gland tissue in BPH rats,reduce the proliferation of fibrous tissue,and repair prostate function.4.Acupuncture can improve the apoptosis rate of prostate cells in BPH rats and promote apoptosis of prostate cells.5.Expression of BCL-2 in acupuncture group of antiapoptotic proteins in prostate tissue is reduced.The expression of apoptosis proteins bax was promoted.It indicated that acupuncture promoted apoptosis of prostate and improve prostate function witch was related to regulating the expression of apoptosis related proteins.
