ObjectiveThe establishment of rat model which mimicking clinical neonatal hypoxic (HIE)is to study of the acupuncture effect on the expression of NeuroD and cellproliferation in rat brain to provide experimental basis for acupuncture treatment ofHIE disease.Methods1. According to the Grojean’s method: establishment of HIE model by putting the SDrats which were born within10-24h, into100%of the nitrogen anaerobic environmentto evaluate the animal model.2. Newborn SD rats were divided into3groups randomly: the control group, thehypoxia group and the acupuncture group. The corresponding acupuncture points ofrats were selected and were stimulated30minutes per day for10days. We study thevarious expressions of NeuroD by Western blot、 immunofluorescencestaining,RT-PCR and the cell proliferation by immunofluorescence of BrdU.Results1. Hypoxia in neonatal rats were showed abnormal neurological manifestations, suchas: restlessness, disturbance of consciousness, accompanied by systemic cyanosis,cerebral cortex in HE staining, anoxia rat brain nerve cell injury.2. Immunofluorescence shows: NeuroD was detected in cerebral cortex and dentategyrus.RT-PCR result shows that the expression of acupuncture group NeuroD mRNAwas higher than that of the control group and the hypoxia group (P<0.01).Westernblot result shows that the average light density of the acupuncture group NeuroD/β-actin was higher than that of the normal group and the hypoxia group (P<0.01). Theaverage protein of fluorescence intensity in acupuncture group NeuroD were higher than that of the normal group and the hypoxia group (P<0.01); the expression ofSVZ DG acupuncture group of BrdU were higher than that of normal group and thehypoxia group (P<0.01).ConclusionsNeuroD mainly expresses in cerebral cortex、 DG and SVZ.Acupuncture induces the up-regulation of NeuroD and may lead to cellproliferation.
