Objective:To observe the mechanism of scalp point-through-point acupuncture on apoptosis in brain tissue of model rats with cerebral hemorrhage.Method:Wistar rats were randomly divided into normal group, model group and acupuncture group, each group according to different time after making model and randomly divided into6h、1d、3d、7d four time points. The rats having been injected autologous blood to creat cerebral hemorrhage model,acupuncture group were given "Baihui Qubin acupoint" treatment. Observe in rats on each time point.(1) To observe the score of neural function defect at different time points in each group of rats;(2) To observe the HE rats brain tissue pathological changes of dyeing under the light microscope; Penumbra apoptosis was detected by TUNEL assay;(3)Immunohistochemistry and Western blot method are used to detect expression of p38MAPK and P-p38MAPK in brain tissue of rats in each group dynamics in the penumbra of cerebral hemorrhage;(4)Expression in brain tissue of rats were detected by immunohistochemical method in Caspase-3, Bcl-2dynamic in the penumbra of cerebral hemorrhage.Results:(1)Score of neural function defect in acupuncture group showed obvious improvement,in phase3d,7d when compared with the model group, the difference was statistically significant (p<0.05);(2)The results of HE staining:compared with the model group, acupuncture group of nerve cell swelling,interstitial edema, neuron number increase.TUNEL test results: compared with the model group, acupuncture group at each time point the number of apoptotic cells were reduced, and there was significant in3d,7d phase difference (p<0.05);(3)Immunohistochemistry,Western blot results: compared with the model group,the brain tissue of acupuncture group at each time point of p38MAPK and p-p38MAPK protein expression levels were significantly lower,the difference was statistically significant(p<0.05);(4)Immunohistochemistry showed:compared with the model group,the brain tissue of acupuncture group at each time point Caspase-3protein expression decreased, Bcl-2protein expression level increased, and there was significant difference in some time points (p<0.05).Conclusion:(1)The scalp point-through-point acupuncture method can reduce the symptoms of nerve function defect of cerebral hemorrhage rats;(2) The scalp point-through-point acupuncture has inhibitory effect on nerve cell apoptosis of secondary brain injury after intracerebral hemorrhage in rats;(3) Scalp point-through-point acupuncture can inhibit the activation of P38MAPK signal transduction pathway which is closely related to cell apoptosis;(4)The scalp point-through-point acupuncture can inhibit the expression of penumbra region of Caspase-3protein of cerebral hemorrhage, promote the expression of Bcl-2protein, to play the role of anti apoptosis.
