Objective: The purpose of this experiment research is to study the therapeutic effect and difference of Yu's scalp point cluster needling,enriched environment intervention and the acupuncture combined with rehabilitation therapy on APP/PS1 transgenic mice.Taking Bcl-2 and Bax,which are closely related to the apoptosis mechanism of Alzheimer's disease cells,as the point of penetra.We used Morris water maze and Western blot to explore the mechanism of acupuncture combined with rehabilitation therapy on Alzheimer's disease at different levels of behavior and apoptosis factors.Methods: The 7-month-old,clean and male APPswe/PS1 d E9 transgenic mice were stochastically divided into four groups: model group,acupuncture group,enriched environment group and acupuncture combined with rehabilitation therapy group,with 9 mice in each group.In addition,9 wild mice of the same sex and age were used as the blank group.The mice in the enriched environment group were placed in the specially arranged environment,and the mice in the acupuncture group were treated with acupuncture every day.Once a day,one day off after 6 days of treatment.The living environment of mice in the group of head point cluster acupuncture combined with rich environment was arranged the same as that in the group of rich environment,and the acupuncture technique was the same as that in the group of acupuncture.Model group and blank group did not receive any treatment.All of the above treatment lasted for 4 weeks.Morris water maze was used to detect the learning and memory ability of mice,and Western blot was used to observe the changes of Bax and Bcl-2 protein expression in the hippocampus of mice.Result:1.The results of mice escape latency were as follows: The escape latency of the blank group was the shortest on the fifth day,and the model group was the longest.Compared with the other three treatment groups,acupuncture combined with rehabilitation therapy group < acupuncture group< enriched environment group;The results of the second quadrant percentage and the times of crossing platform were: the model group is the smallest,the blank group is the largest.Compared with the three treatment groups,enriched environment group < acupuncture group < acupuncture combined with rehabilitation therapy group.2.Western blot showed that the relative gray value of Bcl-2 protein in hippocampus of mice in each group was: the relative gray value of Bcl-2protein expression in the hippocampus of mice in the model group was the lowest,the highest in the blank group,and compared with the other three groups,enriched environment group < acupuncture group < acupuncture combined with rehabilitation therapy group.The results of relative gray value of Bax protein in hippocampus of mice in each group were as follows: the blank group was the smallest,the model group was the largest,acupuncture combined with rehabilitation therapy group < acupuncture group < enriched environment group.Conclusion:1.The spatial learning and memory ability of APP / PS1 transgenic mice can be improved by the combination of scalp cluster needling and rich environment.And it is better than single scalp cluster needling or rich environment treatment.2.The relative expression of Bcl-2 protein can be incresed and Bax protein can be decreased in the hippocampus of APP/PS1 transgenic,and the apoptosis of neurons in the hippocampus can be inhibited. |
