Objective:To establish an abnormal savda disease carrying hepatocellular carci noma animal model according to Uyghur traditional medicine, investigate the impor-tant role and significance of STAT3, Cyclin D1, p21and p53gene during the pr-ogress of originating and developing of abnormal savda disease carrying hepatocell-ular carcinoma. Method:According to the theory of Uyghur traditional medicine, male wistar rats were fed with cold-natured diet under cool dry environment, stimu-lated with chronic and interrupted electric foot shocks, enforced swimming and co-ntrolled movement to establish abnormal savda syndrome rat model in Uyghur med-icine in three weeks, DEN induced20weeks with further to establish abnormal savda disease earring hepatocellular carsinoma model. Observe the morphological ch-anges of hepatic tissue by using of the microscope respectively. Application of tra-nscriptase-polymerase chain reaction(RT-PCR), immunohistochemistry and western-blot were used to detect the expression level of each group liver tissue STAT3, Cyclin D1, p21and p53genes. Result:cancer rate of abnormal savda disease carr-ying hepatocellular carcinoma model group and cancer cell atypia significantly high-er than hepatoc-ellular carcinoma control group; RT-PCR results showed that the expression of STAT3, Cyclin D1, p21and p53gene in abnormal savda disease car-rying hepatocellular carcinoma model group was higher than the normal control gr-oup and compared with the hepatocellular carcinoma control group also increased significantly,immunohistochemistry and western-blot showed that compared with the hepatocellular carcinoma control group and normal control group the expression of STAT3, Cyclin D1, p21and p53in abnormal savda carrying hepatocellular carcin -oma model group was increased. Conclusion:abnormal savda hilit was the acceler-ative factor in the process of establishing abnormal savda disease carring hepatocel-lular carcinoma model. Illustrates the abnormal savda hilit may through influencing STAT3, Cyclin D1, p21and p53gene of hepatocellular carcinoma, promote the occurrence and development of abnormal savda carring hepatocellular carcinoma. |
