Objective:Based on the establishment of Hepatocarcinoma rat model carrying abnormal savda syndrome, to investigate abnormal savda munziq’s effect on Hepatocarcinoma rat model carrying abnormal savda syndrome and its mechanism. Methods:On the basis of Uyghur traditional medicine theory, establishing the abnormal savda syndrome rat model, further to inducing hepatocarcinoma of carrying abnormal savda syndrome rat model using Diethyl nitrosamine (DEN). Samples were prepared from blank group; hepatocarcinoma group; hepatocarcinoma+Abnormal Savda syndrome group; Hepatocarcinoma rat model carrying abnormal savda syndrome group was administrated with Abnormal Savda Munziq low, medium and high (1.5,3.0 and 6.0g/kg) doses goups respectively. Samples were measured by NMR combined with pattern recognition and used to find out the variability serum metabolites by the method of Orthogonal Partial Least-Squares Discriminant Analysis (OPLS-DA). Results:1) Compared to the normal group and model control group, the both pathological and tissue structure changes are more severe in hepatocarcinoma rat model carrying abnormal savda syndrome group; 2) Compared with the normal group, a wide range of amino acids including serum alanine, glycoprotein, citric acid, tyrosine, phenylalanine, acid content are increased, while the concentration level of α-glucose and P-glucose have decreased in serum of abnormal savda syndrome rats with Hepatocarcinoma. Lactic acid and citric acid are increased while glucose are obviously decreased compared with control group, (P<0.05); 3) We were used to find out that the increased concentration level of a-glucose,β-glucose, taurine in ASM low dose group. Together with the serum of rats in the ASM medium dose group also have increased these three. Lactic acid is decreased, while some essential amino acids are increased in medium dose group, there are no significant changes in ASM high dose group. Conclusion:1) The Hepatocarcinoma carring Abnormal savda rat model establishment is stable and reliable.2) These findings indicate that in abnormal savda syndrome rats with Hepatocarcinoma, lipid metabolism-related metabolites are increased, the abnormal savda syndrome may be accompanied with imbalance of amino metabolic dysfunction of the organism, energy metabolism, resulting in increased the accumulation of lactic acid, resulting in the concentration level of serum alanine, glycoprotein, citric acid, tyrosine, phenylalanine, formic acid content were relatively increased, but the level of both a-glucose and β-glucose were decreased, which may potentially be the metabolic biomarkers of abnormal savda hepatocellular carcinoma. ③Abnormal savda Munziq (medium dose) many acid to regulate amino acid metabolism, Glucose metabolism, such as energy metabolism of abnormal savda rat model with hepatocarcinoma by increasing the content of branched-chain amino acids, Accelerated gluconeogenesis to increase energy supply, reduce the accumulation of lactic.
