Objective:To establish abnormal savda disease carrying hepaticcarcinoma animal model till the hepatocirrhosis occur according to uighur traditional medicine,to investigate the role of the two kinds of gene expression levels and endocrin-immune network on abnormal savda disease carrying hepaticcarcinoma animal model occurrence and development. Method:According to the theory of traditional Uyghur medicine,using multi-factor complex role,to establish abnormal savda syndrome rat model in Uyghur medicine, DEN was used furthurly to establish abnormal savda disease carring hepaticcarcinoma animal model till the hepatocirrhos is occur. Observe the morphological changes of hepatic tissue and structure changes of hepatocellular by using of the optic microscope;The serum levels of Adrenocorticotropic hormone (ACTH),Cortisol hormone(CORT) examined by Radioimmunoassay method;Observe morphological structure by optic microscope the morphological structure of Hypothalamic-pituitary-adrenal axis. Detect the altration of IL-1β、IL-6 and TNF-α by using ELISA; Reverse transcription-polymerase chain reaction (RT-PCR) of candidate genes of fatty acid binding protein 4 (FABP4) and cluster of differentiation 24 (CD24) expression in the liver were detected. Results:(1)Microscopic observation show that compared with the model control group, abnormal savda disease group have obvious morphological damages in tissue of the liver.(2) The alterations of ACTH and CORT in serum level were measured. The results show that compared with the normal group the serum levels of ACTH in the model control group and the abnormal savda disease group were increased(P<0.05), compared with the model control group the serum level of ACTH in the abnormal savda disease group were significantly increased(P<0.01);compared with the normal group the serum levels of CORT in the model control group and the abnormal savda disease group were decreased(P<0.05), compared with the model control group the serum level of CORT in the abnormal savda disease group were significantly decreased(P< 0. 05).(3))Microscopic examination show that compared with the normal group,not only model control group, abnormal savda disease group have obvious morphological changes in tissue of the hypothalamus, pituitary, adrenal, but also compared with model control group,abnormal savda disease group morphological changes seriously in hypothalamus, pituitary, adrenal.(4) 20th weeks of DEN induced,compared with normal control group, serum level of IL-1β model control group and abnormal savda disease group were decreased, the results were statistically significant(P<0.05),but compared with model control group, abnormal savda disease group have not significant difference.Compared with normal control group, serum level of IL-6 model control group were decreased, the results were statistically significant(P<0.05),but normal control group, abnormal savda disease group have not significant difference. Compared with normal control group, serum level of TNF-α model control group and abnormal savda disease group were decreased, the results were statistically significant(P<0.05). Compared with model control group, abnormal savda disease group were decreased, the results were statistically significant(P <0.05).(5) The abnormal savda disease group FABP4 mRNA expression level (1.18±0.07),compared with normal control group(0.07±0.009) were significantly increased, and compared with model control group(0.29±0.04) also were increased (P<0.01);The abnormal savda disease group FABP4 mRNA expression level(1.31±0.02),compared with normal control group(0.08±0.003) were significantly increased, and compared with model control group (0.81±0.05) also were increased (P<0.01). Conclusion:High expression of FABP4 and CD24 in liver tissue of abnormal savda disease carring hepatocellular carcinoma rat model may participated the process of of abnormal savda hepatic carcinoma; abnormal savda syndrome, disorder of endocrin-immune network and the interaction between them is the accelerative factor in the process of abnormal savda disease carring hepatic carcinoma model.
