Objective: To establish abnormal savda disease carrying hepaticcarcinoma animal model till the hepatocirrhosis occur according to uighur traditional medicine, investigate the relationship among abnormal savda? disordered endocrin– immune network and DEN to occur hepatocirrhosis in abnormal savda disease carrying hepaticcarcinoma animal model. Method: According to the theory of Uighur traditional medicine establish abnormal savda syndrome rat model in Uighur medicine, DEN was used furthurly to establish abnormal savda disease carring hepaticcarcinoma animal model till the hepatocirrhosis occur. Observe the morphological changes of hepatic tissue and structure changes of hepatocellular by using of the microscope and electron microscope respectively. Analyse the expression of STAT3 In hepatocellular by using RT-PCR and Western-blot. Detect the altration of IL-1β?IL-6 and TNF-αby using Elisa, ACTH and CORT by using RIA in serum level. Result: (1) Compared with the model control group the pathalogical changes and structure changes of hepatic tissue in abnormal savda syndrome model group were significant and developed more quickly. (2) RT-PCR results showed that the expression of STAT3 mRNA in abnormal savda syndrome model group (11th week) (0.81±0.13) is higher than the control group (0.69±0.16) but no significant differences, in the 15th week of DEN inducing, the expression of STAT3 mRNA in abnormal savda syndrome model group (0.84±0.05) is higher than the control group (0. 68±0. 05) (P < 0. 001) , and compared with the model control group (0.74±0. 06) also increased significantly (P<0.01) . (3) in the 11th week of DEN inducing, Western-blot showed that compared with the model control group and control group the expression of STAT3 in abnormal savda syndrome model group was increased. (4) Compared with the control group the serum level of IL-1β?IL-6 and TNF-αin abnormal savda syndrome model group were increased (IL-1β, P<0.01; IL-6?TNF-α, P<0. 001) ; compared with model control group the serum level of IL-6 and TNF-αin abnormal savda syndrome model group were increased (P < 0. 01) ; compared with the control group, the serum level of ACTH, CORT in model control group, abnormal savda disease group, abnormal savda syndrome model group were all increased, and shows significant mathematical differences, compared with model control group the serum level of ACTH in abnormal savda syndrome model group was increased significantly (P < 0.05).Conclusion: Compared with the model control group the hepatocirrhosis in abnormal savda syndrome model group was developed more quickly. Abnormal savda disease increases the process of hepatocirrhosis and its developing by influencing the gene STAT3. The endocrin– immune network in the abnormal savda disease carring hepaticcarcinoma animal model was disoredered till the hepatocirrhosis occur. DEN is the main induction agent, and abnormal savda disease is the accelerative factor in the process of abnormal savda disease carring hepaticcarcinoma model, besides abnormal savda disease ? disorderness of endocrin– immune network?and DEN are closely related and stimulated each other .
