The other 5 healthy control subjects were matched to the cases on gender and age(±2 years). Peripheral blood sample was collected from each participant, 2 samples of each group was detected by whole genome expression chart chip. The differential expression genes related to gastric carcinoma metastasis were analysed by Panther system software and the result of gene expression profile were made a further confirmation by real-time florescence quantitative polymerase chain reaction using the other samples of each group.2 The differential expressions of CTNNB1 and TGFαmRNA in peripheral blood of gastroenteric tumor patients.A total of 126 gastroenteric tumor patients diagnosed with clinical and histological pathology were involved. The living and behavior style of patients was registration with the structured questionnaire. 65 healthy control subjects were as the controls in the same hospital synchronization. About 3 ml blood was collected from each participant and the expressions of CTNNB1 and TGFαmRNA were detected by real-time florescence quantitative polymerase chain reaction. Above the data was analysed by SPSS 11.5 for windows software.[Results]1 157 masccline gene related to gastric carcinoma metastasis consisted of 65 down-regulation genes and 92 up-regulation genes were sieved by chip. The differential expressions of CTNNB1 and TGFαin gastric carcinoma with metastasis were validated by real-time florescence quantitative polymerase chain reaction, and the result showed that the CTNNB1 was no significant change and the TGFαwas up -regulation with the same expression trend as that in microarray.2 There were no significantly different expressions of CTNNB1 and TGF-αmRNA in peripheral blood between gastric carcinoma and colonretal carcinoma cases, as well as preoperative and postop cases, while there were no significantly different expressions of CTNNB1 and TGFαmRNA in peripheral blood between gastroenteric tumor cases and the healthy controls, as well as whether was gastroenteric tumor with lymph nodes metastasis or with distant metastasis were no significant difference.3 The distributions of expressions of CTNNB1 and TGFαmRNA in peripheral blood of gastroenteric tumor cases were obvious skewness. Compared with male cases, the expression level of TGFαmRNA in peripheral blood of female gastroenteric tumor cases was significantly increased (P<0.05),while compared with cases above 50 years, the expression level of TGFαmRNA in peripheral blood of gastroenteric tumor cases below 50 years was significantly increased(P<0.05). The expression level of CTNNB1 mRNA in peripheral blood had no difference between sex and age.4 When taken distant metastasis as a prognostic dependent variable, the independent variable of clinical stage went into regression equation and the best multivariate logistic regression equation is Logit P=-22.084+5.537X1. [Conclusions]1 157 masccline gene related to metastasis consisted of 65 down-regulation genes and 92 up-regulation genes were found by the whole genome expression chart chip. It could provide clues for searching for new gastric carcinoma metastasis molecular marker.2 There were no significantly different expression level of CTNNB1 and TGFαmRNA in peripheral blood between gastric carcinoma and colonretal carcinoma, as well as between gastroenteric tumor with or without lymphnodes metastasis or distant metastasis.
