Purpose:Resistance training has significant effect on reducing fat and increasing muscle,but the effect of resistance training varies among individuals.In this study,by exploring the association between LEP and MC4R gene polymorphisms and resistance training effects,molecular markers that can predict resistance training and improve body composition were screened,so as to provide a reference for the formulation of precise exercise and fitness programs.Methods:Chinese Han ordinary college students(n=191)underwent resistance training intervention for 12 weeks(twice a week),using barbell squat and barbell bench press with a load of 10×5 groups and an intensity of 70%1RM.Body composition indexes(body weight,BMI,waist-to-hip fat ratio,body fat percentage,muscle weight,ultrasonic muscle thickness,ultrasonic fat thickness)and strength test(isokinetic testing,isometric test,squat jump test)were measured before and after intervention,venous blood was collected to extract serum and DNA,and four lipids and glucose were tested.LEP(Leptin)rs2167270,rs12706832,rs11760956 and MC4R(melanocortin-4 Receptor)rs 17782313,rs6567160,rs476828 polymorphisms were detected by gene microarray.One-way ANOVA and nonparametric test were used,and the significance level was set atP<0.05.The association analysis of LEP and MC4R gene polymorphisms with lipid metabolism,body composition and muscle mass related indexes was conducted to analyze the association between the improvement effect of body composition and gene polymorphism,and to screen molecular markers that can predict the effect of resistance trainingResults:(1)There were differences in the effect of reducing triglyceride among different LEP genotypes at rs2167270,rs 12706832 and rs11760956(P<0.05).The effect of reducing triglyceride in A A genotype at rs2167270,rs12706832 and rs11760956 was the most significant,but there was no significant difference in the effect of reducing triglyceride among AAA,GGG and GAG haplotypes(P>0.05).There was no significant difference between genotypes of LEP gene rs2167270,rs12706832 and rs11760956(P>0.05),and there was no significant difference between haplotypes of AAA,GGG and GAG(P BBB>05).(2)There was a significant correlation between MC4R gene rs6567160 locus and the increase of isometric contraction peak strength among different genotypes(P<0.05),and the increase of isometric contraction peak strength among CT genotypes was the most significant;There was no significant correlation between rs17782313 and rs476828 genotypes and the increase of isometric contraction peak strength(P>0.05),and there was no significant correlation between CCC and TTT haplotypes and the increase of isometric contraction peak strength(P>05).There were no significant differences in serum lipidemia and body composition among MC4R gene rs17782313,rs6567160 and rs476828 genotypes,CCC and TTT haplotypes(P>0.05).Conclusion:(1)The AA genotypes at rs2167270,rs12706823 and rs11760956 of LEP genes were relatively sensitive to triglyceride reduction after resistance training.(2)The CT genotype of rs6567160 locus of MC4R gene was relatively sensitive to the increase of muscle strength after resistance training.
