| Objective:The intervention effect of Duhuo JiSheng Decoction on lumbar intervertebral disc rats was observed,providing data and theoretical support for the clinical application of Duhuo JiSheng decoction in the treatment of lumbar intervertebral disc degeneration.Methods:1.Grouping:After 2 weeks of adaptive feeding,the 30 male SD rats were divided into 3 groups according to the random number table,namely blank group,model group,and administration group,and 10 rats in each group.Before modeling,spontaneous pain behavior scores and neurological function judgment scores were performed on each group of rats.2.Modeling:The blank group was given intraperitoneal injection of 0.9%normal saline(2.5ml/kg),and the model group and the administration group used "lumbar posterior column structural resection" to construct IDD rats.The specific method is:first cut the skin tendons and fascia of healthy SD rats layer by layer,resect the upper and lower spinous ligaments and interspinous ligaments and the upper and lower articular joints on both sides of the waist 1~3,and suture the muscles,subcutaneous and skin wounds layer by layer.After surgery,penicillin intramuscular injection was given to the model group and the administration group for 3 consecutive days,80,000 units/d.Spontaneous pain behavior scores and neurological function judgment scores were performed on the 3rd,1st,2nd,3rd and 4th weeks after modeling.3.Administration:On the 3rd day after the end of modeling,gavage will be started from 11 am to 12 am and 5 pm to 6 pm every day,the blank group and the model group will be given the same volume of 0.9%normal saline injection for gastric gavage,and the administration group will be given 3ml/100g of solitary parasitic soup for 8 weeks of continuous gavage.4.Detection:The day after the end of gavage,3%pentobarbital sodium was injected intraperitoneally to carry out anesthesia,and after successful anesthesia,the spine CT test was performed on each group of rats,and imaging analysis was performed,and the materials were taken after the detection.5.Observation indicators:(1)Observe the general condition of each group of rats.(2)Observe the spontaneous pain behavior score and neurological function judgment score at different time points after molding in each group.(3)CT was used to detect the curvature and structural changes of the lumbar spine of rats in each group.(4)The tissue morphology of rats in each group was observed by HE staining.Results:1.General situation:After the end of molding,the vital signs of the model group and the drug group were stable,but the spirit and mobility were poor,and the wounds of the model group and the drug group did not have obvious bleeding and exudation and skin ulceration.Compared with the blank group,the symptoms of lumbar degeneration occurred in the rats in the model group and the administration group,and the symptoms of lumbar degeneration in the rats were improved after the intervention of solitary parasitic decoction in the drug group.2.Behavioral results:(1)The results of spontaneous pain behavior score showed that the rats in each group performed well in pain behavior before modeling.There was no significant difference(P>0.05),after modeling,the spontaneous pain behavior scores of the model group and the drug group were increased at 3 days and 1 week after surgery(P<0.05),at 2 weeks after surgery,there was no significant difference in the spontaneous pain behavior scores between the model group and the drug group(P>0.05),and at the 3rd and 4th weeks postoperatively,the spontaneous pain behavior score of the drug group was reduced compared with the model group(P<0.05).(2)The results of neurological function scores showed that the neurological function scores of rats in each group before modeling were good and there was no significant difference(P>0.05);after modeling,the neurological function scores of the model group and the drug group were increased at 3 days and 1 week after surgery(P<0.05),and the neurological function scores of the drug group were reduced(P<0.05)in the drug group 2 weeks after surgery,and the neurological function scores of the drug group were reduced(P<0.05)in the third and fourth weeks after surgery.3.Lumbar CT performance:The lumbar vertebral space of rats in the blank group was normal,and the curvature of the lumbar spine was acceptable.The vertebral body sequence was complete and normal,there was no osteophyte on the upper and lower endplates of the vertebral body,the cartilage endplate was basically intact,and no obvious calcification was seen,and the lumbar vertebrae of the rats in the model group had narrowing of the intervertebral space,the vertebral body structure was unstable,the curvature of the lumbar spine became straight,and there was osteophyte hyperplasia and obvious calcification.Compared with the model group,the above performance of rats in the drug group was improved.4.HE staining results:The nucleus pulposus cells in the blank group were arranged normally,and no obvious inflammatory cell infiltration was seen,while the nucleus pulposus tissue of the lumbar intervertebral disc in the model group showed a decrease in nucleus pulposus cells,a disordered fiber arrangement,an increase in inflammatory cell infiltration,and vacuolar changes.Compared with the model group,the damaged nucleus pulposus was repaired in the drug group compared with the previous step,and there was a little inflammatory cell infiltration.Conclusion:1.By resecting part of the posterior column tissue of the spine,thereby affecting the stability of the vertebral body as a whole,the instability of the spine can induce and aggravate the degeneration of the intervertebral disc,resulting in changes in morphology and function,and the lumbar intervertebral disc degeneration model can be successfully constructed.2.The solitary parasitic decoction can delay the degeneration of the lumbar intervertebral disc to a certain extent,thereby improving the curvature of the lumbar spine.3.The solitary parasitic decoction can improve pain symptoms and nerve function in rats with lumbar disc degeneration. |
