| [Objective]:By establishing mouse model of perianal abscess surgery wound and intervening it with Xuexiang powder,which is made from the prescription of “the therapy of roasting pus to promote regeneration”,observes the effect of Xuexiang powder on the perianal abscess surgery wound healing.By measuring the relevant indexes of mice,analyzes the possible mechanism of Xuexiang powder in promoting healing of perianal abscess surgery wound,and provides experimental basis for the clinical application of Xuexiang powder,and provides new ideas and choices for the intervention treatment of perianal abscess surgery wound.[Methods]:Forty SD mice were selected and divided into two groups according to random number table,10 in the normal group and 30 in the model group.Feces suspension was prepared from fresh feces of healthy adults.A circular incision was made on the back of each mouse in the model group,and the feces suspension was dropped on it and wrapped with gauze for 48 h to make the model.After successful modeling,according to the random number table method,30 mice were randomly divided into model group,ethacridine lactate group and Xuexiang powder group,with 10 mice in each group.The normal group take a normal diet and without any intervention;In the model group,1 ml of feces suspension was dripped on the wound 0.5 h before dressing change.At the beginning of dressing change,50 ml of normal saline was taken to wash the wound repeatedly,and then the prepared normal saline gauze was applied to the wound,covered with gauze and fixed with medical tape.Change the dressing at 10 am every day until the wound is completely healed;Ethacridine lactate group and Xuexiang powder group were treated with ethacridine lactate gauze and Xuexiang powder gauze respectively,and the other operations were the same as those in the model group.After treatment,the closure index of the wound surface in mice was measured on the 5th,10 th and 15 th day,and the white blood cell count and neutrophil percentage in each group were detected by whole blood cell analyzer on the 1st and 15 th day.On the 15 th day after treatment,all the mice in each model group were taken a tissue which is about 0.5cm wide from the edge to the center of the wound.Three tissues in each group were randomly taken for Elisa to determine the levels of VEGF and b-FGF,and three tissues were used for Elisa to determine the levels of TNF-α,IL-6 and IL-4.Three tissues were used to determine the protein expression of VEGF and b-FGF by Western blot,and one tissue was randomly selected for HE staining to observe the wound recovery of mice in each group(In the normal group,one mouse was randomly selected to take normal tissue from its back for HE staining as a control).Finally,the mice were killed after dressing change every day until the wound healed completely,and the time of wound healing in each group was recorded.[Results]:(1)The complete wound healing time of Xuexiang powder group and ethacridine lactate group was significantly shorter than model group and the difference was statistically significant(P<0.001,P<0.01).The wound healing time of Xuexiang powder group was shorter than that of ethacridine lactate group,and the difference was statistically significant(P<0.01).(2)The closure index of the mice wounds on 5d,10 d and 15 d of the Xuexiang powder group and the ethacridine lactate group were greater than the model group,and the differences were statistically significant(P<0.01).The closure indices of mice wound on the 5d,10 d and 15 d of the Xuexiang powder group were higher than ethacridine lactate group,and the difference was statistically significant(P<0.001).(3)Compared with the normal group,the cytology inflammation indicators on the 1d of the Xuexiang powder group,the ethacridine lactate group and the model group were significantly increased and the differences were statistically significant(P<0.01).There was no significant difference in the cytology inflammation indicators on the 1d between the Xuexiang powder group,the ethacridine lactate group and the model group(P>0.05).Compared with the normal group,the cytology inflammation indexes on the 15 d day of mice in the Xuexiang powder group,the ethacridine lactate group and the model group were higher than that in the normal group,and the differences were statistically significant(P<0.05).Compared with the model group,the cytology inflammation indicators on the 15 d day of mice in the Xuexiang powder group and the ethacridine lactate group were lower than that in the model group,and the differences were statistically significant(P<0.01).The cytology inflammation index on the 15 d day of the mice in the Xuexiang powder group was lower than that in the ethacridine lactate group,and the difference was statistically significant(P<0.05).(4)Elisa was used to test the expression levels of IL-4 in the back wound tissue of mice in the Xuexiang powder group and ethacridine lactate group on the 15 d.The expression levels were significantly higher than model group and the differences were statistically significant(P<0.01).The expression level of IL-4 in the back wound tissue of mice in the Xuexiang Powder group on the 15 d was higher than the ethacridine lactate group,and the difference was statistically significant(P<0.001).The expression levels of IL-6 and TNF-α in the back wound tissue of mice in the Xuexiang powder group and ethacridine lactate group on the 15 d were significantly lower than those in the model group and the differences were statistically significant(P<0.01).The expression levels of IL-6 and TNF-α in the back wound tissue of mice in the Xuexiang powder group on the 15 d were lower than those in the ethacridine lactate group,and the differences were statistically significant(P<0.01).(5)Elisa showed that the expression levels of VEGF and b-FGF in the back wound tissue of mice in the Xuexiang powder group and ethacridine lactate group on the 15 d were higher than those in the model group and the differences were statistically significant(P<0.01,P<0.001);The expression levels of VEGF and b-FGF in the back wound tissue of mice in the Xuexiang powder group on the 15 d were higher than those in the ethacridine lactate group,and the difference was statistically significant(P<0.001).(6)Western blot analysis showed that the protein expressions of VEGF and b-FGF in the back wound tissue of mice in the Xuexiang powder group and ethacridine lactate group on the 15 d were significantly higher than those in the model group and the differences were statistically significant(P<0.01).The protein expression levels of VEGF and b-FGF in the back wound tissue of mice in the Xuexiang powder group on the 15 th day were higher than those in the ethacridine lactate group,and the difference was statistically significant(P<0.05).(7)HE staining results of local tissues of wounds on 15 d after modeling in the Xuexiang powder group,the ethacridine lactate group and the model group showed that the overall structure of the wound tissues in the Xuexiang powder group was basically normal as compared with those in the later two groups,with visible hair follicles and dermis,no significant rupture,atrophy and swelling of collagen fibers,and thickening of the muscular layer.[Conclusion]:(1)By making a suitable wound on the back of mice,adding normal human fecal suspension can successfully mold the postoperative wound model of perianal abscess that meets the experimental requirements,and it is easy to operate and easy to perform.(2)The efficacy of Xuexiang powder in alleviating wound inflammation,shortening wound healing time and improving wound healing quality of perianal abscess surgery wound mice model was superior to ethacridine lactate.(3)Xuexiang powder can reduce wounds and systemic inflammation in mice by affecting the levels of anti-inflammatory and pro-inflammatory factors such as IL-4,IL-6,and TNF-α in the model mice of perianal abscess surgery wound.(4)Xuexiang powder can shorten the time of wound healing and improve the quality of wound healing by up-regulating the expression of VEGF and b-FGF in the model mice of perianal abscess surgery wound.(5)Xuexiang powder,which is the combination of the prescription,guided by “the therapy of roasting pus to promote regeneration”,can promote perianal abscess surgery wound healing by up-regulating the expression of VEGF and b-FGF. |
