| Objective: To explore the relationship between liver and kidney health and disease in Mongolian medicine,to clarify the relationship between the two kidneys and the pharmacological action of the prescription for cold syndrome of liver and kidney.Methods: SPF 200 SD male rats were randomly divided into 12 groups after feeding for one week.The rats were randomly divided into normal group,model group and treatment group.(1)normal group(2)adriamycin model group(3)kidney cold model group;(1)The model of renal cold syndrome was established by applying ice pack to the dorsal renal area of rats for 6hours a day for 45 days.(2)The right kidney of SD rats was injected with adriamycin to make the right kidney lose renal function,but the left kidney was normal.Chronic nephropathy of the right kidney was replicated.Then,the rat’s left kidney was cold applied with ice pack for 6hours a day for 45 days to establish the model of kidney cold syndrome.(3)The left kidney of SD rats was injected with adriamycin to make the left kidney lose kidney function,but the right kidney was normal.Then cold compress the dorsalright kidney of the rats with ice pack for 6 hours a day for 45 days to establish the model of kidney cold syndrome.After the establishment of the kidney cold model,the patients were randomly divided into model group,Mongolian Medicine Yizhi Shiwei SAN and Errigna Anari-8.Mongolian medicine was used for intervention,and the patients were given intragastric administration once a day for 30 days.cAMP,cGMP,Renin,Cort,IL-27,IL-6,IL-1,IL-2,MCP-1,IL-1α,IL-1β,TNF-α,IL-8,Heparin,Urea,Cre,ALT,AST,ALB,ALP and TP were detected in the blood samples.cAMP,cGMP,IL-27,IL-6,Renin and Cort were detected in left and right kidney tissue samples.cAMP,cGMP,IL-27,IL-6,Heparin,Renin and Cort were detected in liver tissue samples.The protein expression of TGR5 in left and right kidney and liver tissues and the efficacy of Mongolian medicine were detected by Westernblot.The protein expression levels of Renin and Cort in left and right kidney and liver tissues and the efficacy of Mongolian medicine were detected by immunohistochemical method,and the mechanism of action of kidney cold syndrome on kidney and liver and the relationship between normal kidney and liver and disease as well as the relationship between two kidneys were preliminarily discussed.Pathological changes of kidney and liver tissues and the efficacy of Mongolian medicine were observed by HE staining method.The above indexes were used to investigate the effect of kidney cold syndrome on liver function and the prevention and treatment effect of Mongolian medicine.Results: Kidney cold syndrome was induced by applying cold and wet ice pack to the dorsal renal area of rats.The typical manifestations of kidney cold syndrome appeared,such as cold limbs,frequent and white urination,sparse food and loose stool,curled limbs,and serum cAMP level and cAMP/cGMP ratio were significantly reduced.cAMP level increased significantly,cGMP level decreased significantly and cAMP/cGMP ratio increased significantly.cAMP level and cAMP/cGMP ratio were significantly increased in liver tissue.The levels of IL-6 and renin in the left kidney were significantly increased,while the levels of IL-27 and cortisol were significantly decreased.The levels of IL-6 and IL-27 in the right kidney were significantly decreased,while the levels of renin and cortisol were significantly increased.The levels of IL-6,heparin,renin and cortisol in liver tissue increased significantly,while the levels of IL-27 decreased significantly.The levels of androgens and estrogens in the left kidney and liver were significantly reduced.These results support the successful modeling of kidney cold syndrome in this study.Renal cold syndrome can significantly increase the serum levels of IL-27,IL-6,IL-1,MCP-1,IL-1α,IL-1β and TNF-α,cause inflammation and apoptosis,lead to decreased renal function,significantly increase the blood urea nitrogen content and significantly decrease the blood creatinine content.After 45 days of cold compress in both normal kidneys,Cort protein expression in left kidney caused by renal chill was not significantly different,while Cort protein expression in right kidney and liver was significantly decreased,while Renin protein expression in left kidney and liver was significantly increased,but there was no significant difference in right kidney.There was no difference in the expression of TGR5 protein in the left and right kidneys and liver.Doxorubicin was injected into the left kidney to make the left kidney lose renal function and the right kidney normal.Then cold compress was applied to the dorsal right kidney of the rats for 45 days,the expression of Cort protein in the right kidney and liver caused by renal chill was significantly decreased,and the expression of Renin protein in the liver was significantly increased,while the expression of Renin protein in the right kidney was not significantly different.There was no difference in the expression of TGR5 protein in right kidney and liver.Doxorubicin was injected into the right kidney to make the right kidney lose function and the left kidney normal,and then cold compress the dorsal left kidney area of the rat for 45 days.There was no significant difference in the expression of Cort and Renin protein in the left kidney caused by renal chill,and the expression of TGR5 protein was significantly decreased,while the expression of Cort protein was significantly decreased and the expression of Renin protein was significantly increased in the liver,and the expression of TGR5 protein in the liver was not significantly different.In normal condition,the concentration of cAMP,cGMP and cAMP/cGMP in the left and right kidneys were different.Compared with the normal left kidney,the level of cAMP in the normal right kidney was significantly increased,the level of cGMP was significantly decreased,and the ratio of cAMP/cGMP was significantly increased.There was no significant difference in cAMP,cGMP and cAMP/cGMP ratio of left and right kidneys after 45 years of cold compress in the dorsal renal area of rats.Further studies showed that the levels of IL-6 and Renin in the normal right kidney were significantly increased,while the levels of IL-27 and Cort were significantly decreased compared with the normal left kidney.After 45 years of cold compress on the dorsal renal area of rats,IL-27 in the right kidney of the model was significantly decreased compared with the left kidney,while IL-6,Renin and Cort were not significantly different.There was no significant difference in the levels of androgens and estrogens in the left and right kidneys between normal and diseased states.In this study,compared with normal liver,cAMP and cGMP levels in left kidney were significantly decreased,while cAMP/cGMP levels were not significantly different,cAMP level in right kidney was significantly increased,cGMP level was significantly decreased,and cAMP/cGMP level was significantly increased.Compared with model liver,cAMP,cGMP,cAMP/cGMP ratio of model left and right kidneys had no significant difference.Compared with the normal liver,the Cort level in the normal left kidney was significantly increased,the Renin level was decreased,the IL-6level in the normal right kidney was significantly increased,and the IL-27 and Cort levels were significantly decreased.Compared with model liver,IL-27 level in model left kidney was significantly increased and Cort level was significantly decreased.The Cort level in the right kidney of the model was significantly decreased.Mongolian medicine Yizhi Shiwei Powder has the functions of dispelling kidney cold,improving kidney function and diuresis,etc.Mongolian medicine Erriganari-8 has the functions of dispelling liver cold,warming liver,regulating qi and assisting digestion [19].The results of this study show that Mongolian medicine Yizhi Shiwei powder can effectively improve the high urea caused by kidney cold,adjust the physiological function state of the body,the therapeutic effect is satisfactory,and has the value of clinical promotion.Conclusion: The relationship between kidney and liver is "contradictory" in the normal state,but the relationship is unified in the disease state.The function and function of the two kidneys are different;Yizhi Shiwei Powder and Erriganari-8have certain effect on kidney cold syndrome;An external injection of adriamycin can establish chronic kidney disease. |
