Objective:to access effects of herbs tonifying the Kidney on submaxillary-testis axis and to evaluate the role of submaxillary-testis axis in the theory,which the Kidney governs the reproduction by experiments of Kidney-deficiency mice and sialoadebectemized mine. Methods:Some mice were made into Kidney-deficiency and spermatogenesis dysfunction models by given adenine orally,and some mice were made into EGF-deficiency and spermatogenesis dysfunction models by submaxillary gland resection. Volumes of mature spermatozoa in epididymis and spermatogenic cells in seminiferous tubules were examined after experimental mice had been given herbs tonifying the Kidney for one spermatogenic cycle. Expressions of epidermal growth factor receptors (EGFR) and Ki-67 antigen in seminiferous tubules were analyzed by immunohistochemistry and image analysis system,and secretion of granules in submaxillary intercalated ducts was examined by Toluidine Blue staining. Results:Kidney-deficiency and spermatogenesis dysfunction mouse have signs of Kidney deficiency and spermatogenesis dysfunction,while concentrations of serum testosterone and EOF were increased,proliferation of spermatogenic cells in seminiferous tubules was promoted,as well as expression of epidermal growth factor receptors and Ki-67 antigen when herbs tonifying the Kidney were given. Decrease of serum EOF and spermatogenesis dysfunction were founded in sialoadebectemized mouse,while concentration of serum EOF and volume of mature spermatozoa in epididymis were increased,as well as and proliferation of spermatogenic cells in seminiferous tubules after given herbs tonifying the Kidney. There is a correlation between volume of mature spermatozoa in epididymis,serum testosterone and EOF and EGFR expression in seminiferous tubules. Conclusion:Herbs tonifying the Kidney has an effect on submaxillary-testis axis,increasing concentration of serum EGF,which plays a role in spermatogenesis?indicating that submaxillary-testis axis has a relationship with the theory that the Kidney governs the reproduction.
