Objective:Based on 16 Sr DNA high-throughput sequencing technology,this experiment will study the correlation between intestinal flora and PCOS,explore the difference of intestinal flora between phlegm-dampness PCOS patients and normal people,and analyze the difference.To explore the role of intestinal flora imbalance in the pathogenesis of PCOS,and to find a breakthrough point for the treatment of PCOS with traditional Chinese medicine.Methods : Collected from February 2022 to February 2023 in xinjiang selected in the fourth affiliated hospital of xinjiang medical university of traditional Chinese medicine gynecology clinic is phlegm dampness type PCOS patients,medical history collection and physical examination,complete clinical laboratory,auxiliary examination.At the same time,the normal population of gynecological clinic was selected.Based on 16 sr DNA technology,the V3-V4 region of 16 S r DNA was amplified by PCR and sequenced using Illumina PE25 platform.PCo A method based on unweightde_Unifrac distance was used to analyze and compare β diversity.LEf Se analysis method was used to compare the results of intestinal flora in each group,and the structural characteristics of intestinal flora were analyzed.KEGG function prediction was used to predict the pathway of the whole community and obtain the functional information of the sample.Results : 1.(1)This study included 30 subjects(Health group,TS.PCOS group,FTS.PCOS group).There was no significant difference in age,BMI,P,FSH and other general data among the three groups(P > 0.05).There were significant differences in WHR,E2,LH and T among the three groups(P < 0.05).After LSD pairwise comparison analysis,it can be seen that the levels of WHR and LH in the phlegm-dampness PCOS group and the non-phlegm-dampness PCOS group were significantly higher than those in the healthy group(P < 0.05).The E2 level in the phlegm-dampness PCOS group was significantly higher than that in the non-phlegm-dampness PCOS group(P < 0.05).The T level in the non-phlegm-dampness PCOS group was significantly higher than that in the healthy group(P < 0.05).2.There was no significant difference in Chao1 index,Observed species index and Goods coverage index among the three groups(P > 0.05),and there were significant differences in Shannon index and Simpson index(P < 0.05).The PCo A method based on unweightde_Unifrac distance was used to compare the distribution of flora among the three groups.The phlegm-dampness PCOS group(TS.PCOS)and the non-phlegm-dampness PCOS group(FTS.PCOS)were closer,and the similarity of intestinal flora between the two groups was higher;the distance from the healthy group(Health)is far,indicating that the difference between the PCOS group and the healthy group is high,that is,the richness is different.The ANOSIM analysis based on the Unweighted-unifrac distance algorithm showed that the R value was 0.165 and the P value was 0.003,indicating that the difference between the groups in this experiment was large.Conclusion : 1.There are significant differences in intestinal flora between PCOS patients and healthy women.2.There are differences in functional metabolic pathways between the intestinal flora of phlegm-dampness PCOS patients and non-phlegm-dampness PCOS and healthy people,among which the differences in circulation,cardiovascular system and chronic inflammation are the most significant.3.LEf Se analysis showed that Firmicutes could be used as a biomarker for PCOS population in phlegm-dampness and non-phlegm-dampness PCOS groups. |
