Objective: This study mainly investigated the association between AHNAK2 gene single nucleotide polymorphisms and coronary artery disease in Chinese Xinjiang Uyghur and Han populations. Methods: This study used a case-control design and selected 1212patients(602 Uyghur and 610 Han)who were hospitalized and underwent coronary angiography at the Heart Center of the First Affiliated Hospital of Xinjiang Medical University from January 2019 to December 2021.Based on the angiography results, the patients were divided into the CAD group(301 Uyghur and 305 Han)and the control group(301 Uyghur and 305 Han). The i MLDR technique was used to genotype the AHNAK2 gene rs12882641,rs28583515,and rs2582497 loci, and the association between AHNAK2 gene polymorphisms and coronary artery disease was analyzed Results: In the Uyghur population, there were significant differences(P<0.05)in the allele frequency and dominant model distribution frequency of the rs2582497 locus between the CAD group and the control group. The rs28583515 locus showed significant differences(P<0.05)in the genotype distribution, allele frequency, dominant model, recessive model, and additive model distribution frequency between the two groups, and was associated with coronary artery disease in the Uyghur population of Xinjiang. After adjusting for confounding factors, binary multivariate logistic regression analysis showed that the association between rs28583515 locus and coronary artery disease remained significant. The distribution frequency of the G-T-C haplotype(H5)constructed from the rs12882641,rs28583515,and rs2582497 loci was significantly higher in the CAD group than in the control group. In the Han population, there were significant differences(P<0.05)in the distribution frequency of the recessive model and allele frequency of the rs12882641 and rs28583515 loci between the CAD group and the control group. The rs2582497 locus showed significant differences(P<0.05)in the genotype distribution, allele frequency, dominant model, and recessive model distribution frequency between the two groups, and was associated with coronary artery disease in the Han population. After adjusting for confounding factors, binary multivariate logistic regression analysis showed that the association between rs2582497 locus and coronary artery disease remained significant. The distribution frequency of the A-C haplotype(H1)constructed from the rs12882641 and rs28583515 loci was significantly higher in the control group than in the CAD group, while the distribution frequency of the G-T haplotype(H4)was significantly higher in the CAD group than in the control group. Conclusion: This study suggests that the AHNAK2gene’s rs28583515 locus is associated with coronary artery disease in the Chinese Uyghur population in Xinjiang, while the rs2582497 locus is associated with coronary artery disease in the Han population. The G-T-C haplotype(H5)constructed by rs12882641,rs28583515,and rs2582497 loci may be a risk factor for coronary artery disease in the Uyghur population in Xinjiang. In the Han population, the A-C haplotype(H1)constructed by rs12882641 and rs28583515 loci may have a protective effect against coronary artery disease, while the G-T haplotype(H4)may be a risk factor for coronary artery disease in the Xinjiang Han population.
