Objectives To investigate the damage and tissue recovery of low-temperature plasma radiofrequency ablation under different temperatures of 0°C,4°C and 25°C normal saline,in order to provide the most suitable temperature conditions for relevant patients.Methods A total of 36 New Zealand white rabbits with a total of 72 oral buccal mucosa were randomly divided into two groups:long-term observation group and immediate death group.Each 36-sided oral buccal mucosa was randomly divided into three temperature subgroups of 0°C,4°C and 25°C,with 12 sides of oral buccal mucosa each.After the long-term observation group used plasma radiofrequency ablation system and supporting plasma knife to ablate the buccal mucosa on both sides of the New Zealand white rabbit with normal saline at different temperatures as the medium,10 recording points were intelligently analyzed(wound calculation method based on connected domain external contour and statistical ratio filtering)immediately after surgery,day 1,day 3,5 days,7 days,9 days,11 days,13 days,15 days,and 17 days,and the direct injury and overall damage area of the surgical area were compared at three temperatures.The change in injury area,basic healing,and days required for complete healing was assessed at two adjacent recording time points in the surgical area.In the immediate execution group,plasma ablation of the buccal mucosa of the rabbit was immediately sacrificed,and the materials were collected and pathological analysis was performed after the sacrifice to evaluate and compare the depth of tissue damage and the degree of infiltration of inflammatory cells(macrophages and neutrophils)at the three temperatures.Finally,statistical software was used to analyze the ANOVA,T-test or rank sum test on the data of the three temperature subgroups and draw corresponding conclusions.Results(1)In the long-term observation group,the time required for complete healing in the 0°C,4°C and 25°C groups was 12.42±1.11 days,15.17±1.34 days and14.67±1.65 days,and the time required for complete healing in the 0°C group was shorter and statistically different(P<0.05)than that in the 4°C group and 25°C group,but there was no difference between the 4°C group and the 25°C group.The basic healing time of the 0°C,4°C and 25°C groups was 9.08±1.19 days,9.00±1.22 days and9.63±1.03 days,and there was no significant difference in the basic healing time(P>0.05).The total injury area on the third day after surgery in the 0°C,4°C and 25°C groups was 51.68±8.61mm~2,32.29±7.81mm~2 and 23.25±4.38mm~2,and the overall injury area on the 7th day after surgery was 26.06±7.44mm~2,20.55±3.63mm~2 and 16.16±2.48mm~2.The total damage area on day 3 of the 0°C group was greater than that of the 25°C group and the difference was significant(P<0.05),and the total damage area on day 7of the 4°C group was greater than that of the 25°C group and all were statistically significant.The immediate postoperative injury areas of the 0°C,4°C and 25°C groups were 18.33±1.18mm~2,16.30±1.52mm~2 and 16.29±3.12mm~2,and the direct injury areas on the 9th day after surgery were 0.66±0.66mm~2,0.93±0.51mm~2and 1.13±0.36mm~2.The immediate postoperative direct injury area in the 0°C group was greater than that in the 25°C group and was statistically significant.The area of direct injury on day 9 of the 0°C group was lower than that of the 25°C group and was statistically significant.The overall injury area difference from the first day to the third day after surgery in the 0°C,4°C and 25°C groups was-23.16±9.26mm~2,-8.00±7.85mm~2 and10.74±7.21mm~2,and the overall injury area difference from day 7 to day 9 was-4.16±6.04mm~2,13.76±4.09mm~2 and 0.37±1.56mm~2,and the overall injury area difference from day 9 to day 11 was 10.34±5.66mm~2,-0.81±1.58mm~2 and5.22±1.06mm~2,the overall injury area difference from the 11th to the 13th day after surgery was 3.33±2.30mm~2,5.96±1.76mm~2and 1.77±0.63mm~2,and the difference of direct injury area from day 9 to day 11 after surgery was 0.32±0.32mm~2,0.74±0.51mm~2and 0.89±0.30mm~2,and negative values indicated that the damage area of the latter time point increased compared with the previous time point.The recovery of the overall injury from day 1 to day 3 of the 0°C group was worse than that of the 25°C group,the recovery of the overall injury from day 7 to day 9 of the 4°C group was better than that of the 0°C group and the 25°C group,the recovery of the overall injury from day 9 to day 11 of the 4°C group was worse than that of the 25°C group,and the recovery of the overall injury from day 11 to day 13 of the 0°C group was worse than that of the 4°C group.The recovery from direct injury from day 9 to day 11 in the 0°C group was inferior to that in the 4°C group,and the difference was statistically significant.(2)In the immediate death group,the postoperative tissue injury depth of 0°C,4°C and 25°C group was 216.10±22.65mm,265.56±21.10mm and 256.68±51.98mm,and the postoperative tissue injury depth in the 0°C group was significantly shallower and statistically different(P<0.05)compared with the 4°C group and 25°C group,while there was no obvious difference between the tissue damage depth of the 4°C group and the 25°C group.Moderate exudation was 1 case,a small amount of exudation was 5cases,no exudation was 6 cases,the exudation accounted for 50%,the inflammatory cell infiltration in the injury area of the 4°C group was 1 case of large exudation,2 cases of moderate exudation,5 cases of small exudation,4 cases of no exudation,and the proportion of exudation was 66.7%,and the infiltration of inflammatory cells in the injury area of the 25°C group was 0 cases of large exudation,0 cases of moderate exudation,4 cases of small exudation,and 8 cases of no exudation,and the proportion of exudation was 33.3%,and there was no obvious difference between the three groups(P>0.05).The proportion of collagen fibers in postoperative tissue in the 0°C,4°C and25°C groups was 26.54±1.89%,26.28±1.40%and 24.01±1.26%,respectively,and there was no significant difference between the three groups(P>0.05).Conclusion After low-temperature plasma radiofrequency ablation,from the perspective of long-term recovery,the 0°C group has obvious advantages over the 4°C group and the 25°C group in the time of complete recovery of mucosal tissue.There were no significant differences in injury area and healing rate between the three temperature groups.From the perspective of short-term damage,although there was no obvious difference in the degree of inflammatory cell infiltration in a certain area of the three temperature groups,the depth of damage in the 0°C group was significantly lower than that of the other two groups,which had obvious adva-ntages,which meant that lower temperature was more conducive to the prognosis of patients,provided theoretical basis and guarantee for better prognosis of patients,and provided practical diagnosis and treatment reference for the majority of clinicians.
