Low temperature plasma ablation and radiofrequency ablation have the advantages of simple operation,safe reliability and fast recovery,and have been widely used in the treatment of various surgical areas such as otolaryngology,spine surgery and gynecology.At present,low temperature plasma ablation equipments and radiofrequency ablation equipments used by domestic hospitals are mainly imported,and they are expensive,so in order to reduce cost,the research and development work of the two kinds of equipments have gradually carried out in domestic.However,the biological effects of these two methods of ablation and the mechanism of low temperature plasma ablation are not clear.Therefore,the paper carried out the circuit design of low temperature plasma ablation equipment,and carried out a systematic research for the two types of ablation on the ablation effect and temperature effect,which provides important theoretical basis and method guidance for clinical doctors to develop treatment plans and research and development for equipment localization.Specific research contents are as follows:(1)Development of low-temperature plasma surgery system: the design of circuit is mainly DC-DC voltage regulating circuit,DC-AC inverter circuit and high-frequency current sampling circuit.The output voltage is adjustable,overcurrent protection and output current feedback control are realized.(2)Ablation effect of low temperature plasma ablation and radiofrequency ablation : use pork and pig liver as experimental materials,and use low temperature plasma ablation equipments and radiofrequency ablation experiment to do ablation experiments under different voltage,by observing the focal ablation forms and measuring effective ablation area size to analyze the ablation effect of low temperature plasma ablation and radiofrequency ablation for different tissue under different voltage.The results showed that the effective ablation area of low temperature plasma ablation and radiofrequency ablation increased with the increase of voltage.The focal ablation forms differences of low temperature plasma ablation for pork and pig liver were relatively large,but the differences of radiofrequency ablation for pork and pig liver was similar.The effective ablation area of low temperature plasma ablation and radiofrequency ablation of pork and pig liver was changed from no significant difference(P>0.05)to significant difference(P<0.05).(3)Temperature effect of low temperature plasma ablation and radiofrequency ablation : use pork,pig liver and pig fat as experimental materials,and use low temperature plasma ablation equipments and radiofrequency ablation experiment to do ablation experiments under different voltage,by using dynamic thermal analysis system to monitor temperature change in real timeduring ablation,and carry out temperature detection method study,and establish a quantitative analysis method for temperature effect,to analyze the temperature effect of low temperature plasma ablation and radiofrequency ablation for different tissue under different voltage.The results showed that the highest surface temperature,the maximum of the highest surface temperature and the area of thermal damage increased with the increase of the voltage in the process of low temperature plasma ablation and the radiofrequency ablation.Different tissue in the process of low temperature plasma ablation,the maximum of the highest surface temperature and the area of thermal damage of pig fat is significantly smaller than pork and pork liver(P < 0.05),but the temperature effect of pork,pork liver of low temperature plasma ablation and the the temperature effect of three kinds of tissue of radiofrequency ablation is not obvious.(4)Low temperature plasma ablation mechanism: use BSA protein solution and tissue cells as experimental material,and use plasma treatment under different voltage and time,through SDS-PAGE electrophoresis and flow cytometry instrument combining PI fluorescence staining method to preliminarily explore low temperature plasma ablation mechanism.The results showed that during the plasma ablation process,plasma protein molecular chains are indeed play a role in interrupt,meanwhile would destroy the cell membrane structure,and increase the permeability of cell membrane,and tissue cells break down and die,producing cell fragments;The higher the voltage and the longer the time,the more small molecular weight fragments are interrupted,and the higher the proportion of dead tissue cells,the more cells are cracked.
