| Objective:Experiments were conducted to explore the effects of electroacupuncture on the behavior and morphology of rats with slow transit constipation,revealing the principle of electroacupuncture reducing the over-expression of p-ulk1 protein,inhibiting the over-autophagy of icc in damaged colon tissues,increasing the number of icc cells,and reducing the dysfunction of colon transmission after constipation.Method:The experiment was divided into two partsExperiment 1: To study the effect of electroacupuncture on the behavior and histopathology of damaged colon tissue in STC rats.Twenty-four clean SD rats were randomly divided into blank group,model group,electroacupuncture group and drug group,with 6 rats in each group.In the electroacupuncture group,"Zhongwan" point,"Tianshu" point and "Guanyuan" point were selected according to the "Experimental Zoology" and the principle of comparison between experimental animals and human pointsExperiment 2: The expression of p-ulk 1 protein in colon tissues of each group.Twenty-four clean-grade SD rats were randomly divided into 6 blank,model,drug and electroacupuncture group.The preparation model and the intervention method are identical to those used in Experiment 1.The expression of p-ulk 1 protein content in the colon tissues was determined by immunohistochemistry.Results:1.experimental intervention;model group,electroacupuncture group and drug group had dark hair after film making,some rats had loose hair,dry stool,hard texture,long or elliptical shape,irritable temperament and low coordination.Compared with the model group,in the electroacupuncture group and drug group,the stool texture was soft,hair gloss improved,impatience improved and coordination increased.2.The discharge time of the first black stool: compared with the discharge time of the first black stool in the blank group,the discharge time of the model group,drug group and electroacupuncture group was significantly longer.The difference was statistically significant(P<0.01);compared with the model group,the difference was significantly shorter and statistically significant(P<0.01);the difference was statistically significant(P <0.05);compared with the drug group(P<0.01).3.Stool water content: compared with the blank group,the difference was statistically significant(P < 0.01);compared with the model group,the difference increased statistically significant(P< 0.01),and statistically higher(P<0.01);compared with the electroacupuncture group,the fecal water content decreased statistically significant(P<0.01).4.Small bowel propulsion rate: compared with the blank group,the model group was significantly lower and the difference rate was statistically significant(P<0.01);compared with the model group,the difference was statistically significant(P<0.01),and the electroacupuncture group was statistically higher(P<0.01);compared with the electroacupuncture group,the difference was not statistically significant(P<0.01).5.HE results: HE staining results showed that the complete structure of the colon in the blank group was visible,and the mucosal layer,submucosa,muscle layer,muscle layer and outer membrane layer were significantly stratified.The single columnar epithelium of the mucosal layer was arranged more neatly and tightly,with no obvious degeneration and necrosis.A large number of colorectal glands in the lamina propria are single tubular and contain more goblet cells.The submucosa,muscle layer and outer membrane were tightly connected,and no significant pathological changes were observed.No obvious pathological changes were observed;in the colon,the mucosal layer,submucosal layer,muscle layer and outer membrane layer in the model group,small focal accumulation of lymphocytes or neutrophils in the lamina propria;in the drug group,slight inflammatory cells in the native layer,which was closely connected with the outer membrane;in the electroacupuncture group,no significant degeneration and necrosis,and mild lymphocyte or neutrophil infiltration in the lamina propria were observed.Th e submucosa and myometrium were closely connected to the outer membrane with no obvious pathological changes.There are no other obvious pathological changes.6.Immunohistochemistry results showed that P-ulk 1 protein expression was significantly increased in the model group compared with the blank group(P<0.01).P-ulk 1 protein expression was significantly reduced in the drug and electroacupuncture groups compared with the model group(P<0.01).Pulk 1 protein expression was decreased compared to the drug group,but the difference was not significant(P<0.01).Conclusion:1.Electroacupuncture can improve colon transmission function and promote the recovery of colon transmission function in STC rats.2.Electroacupuncture can reduce the over-expression of phosphorylated autophagy associated protein 1(P-ulk1)in colon tissue,reduce the overautophagy of icc in colon tissue of STC rats,increase the number of icc cells,promote colon transport,and thus improve constipation symptoms. |
