Objectives To evaluate the mechanism of Yiqi jianpi tongbian fang that can cure rats with slow transit constipation(STC),which may refer the PI3K/AKT/e NOS signaling pathway in cajal cells of colon tissue.STC is a kind of Functional constipation(FC),is refers to the dysfunction of colonic transit,the jogging speed of the contents of the colon and the colon shrank.Yiqi jianpi tongbian fang is Chinese medicine compound preparation,which has obvious curative effect on patients with slow transit constipation in china.Methods 72 healthy SD rats were randomly divided into blank control group,model group and moshabili group,18 each group.In each group,the rats were administered with morphine(2.5mg/kg)daily to develop STC model after 45 days.Then,all rats were treated with gavage administration: the rats in blank control group were treated with saline,2 times a day,1m L each time;the rats in moxapalili group treated with moxapelli divided into 100 ml distilled water per 15 mg,twice daily,1m L per time;the rats in Yiqi jianpi group were treated with Yiqi jianpi tongbian Chinese medicine,0.75ml/100g/d.Each group of 18 rats was free to eat and drink during the other time.The number and the weight of fecal granule and the body weight of rats were recorded every 5 days.Stop for one week after 4 weeks,transit functions of gastrointestinal movement were examined by an activated charcoal suspension pushing test.The expression of c-kie m RNA in colon cells was detected by PCR.The content of NO and e NOS were detected by ELISA assay.P-Akt expression in colon tissue was observed by western blotting;the expression of cajal cells in the colon tissues was observed with immunohistochemistry.Results 1.Compared with the control group,the average daily fecal granules of every model group were significantly increased,and the average amount of first excretion granule was significantly reduced(p< 0.05)and the number of cajal cells in the colon tissue of every model group was significantly increased.2.Compared with the control group,the contents of NO and e NOS in the colon tissues of model group rats were significantly reduced(p< 0.05).While,the contents of NO and e NOS in the colon tissues of the rats between in mosa bili group and yiqi jianpi group have not statistically significant difference(p>0.05).3.The expression of P-Akt in the colonic in the model group was significantly increased compared with control group(p<0.05).While,the expression of P-Akt the colon tissues of the rats between in mosa bili group and yiqi jianpi group have not statistically significant difference(p>0.05).Conclusions The PI3K/AKT/e NOS signaling pathway is likely to be involved in the mechanism of STC treatment with Yiqi jianpi tongbian fang,and the main target cells that can regulate intestinal peristalsis and transmission by Yiqi jianpi tongbian fang are cajal cells in the colon tissue.
