Abstract/Summary:
BackgroundThroat carcinoma is one of the most common head and neck tumors,accounting forabout a quarter of the total number of head and neck tumors including laryngeal squamous cell carcinoma(LSCC)and accounted for 90% of laryngeal malignant tumor,male incidence is higher than women,the main clinical treatment strategy is surgical resection combined with radiationchemotherapy,but most of LSCC patients still have the risk of recurrence,5 years survival rate is not more than 60%,for advanced LSCC patients treatment effect is not as expected,the prognosis is still very poor.Therefore,it is of great significance to find molecular markers in the process of invasion and LSCC metastasis for the early diagnosis and non-surgical treatment of laryngeal squamous cell carcinoma.The dynein axonemal heavy chain 17(DNAH17)gene,located on chromosome17q25.3,is a large gene composed of 81 exons that encodes the axonemal dynein heavy chain of ODA.The protein encoded by DNAH17 is a subunit of axonemal dynein and is the basic structure of the primary cilia.Many studies have shown that primary cilia play an important role in embryonic development,cell differentiation,cell division,and tumor progression through Hedgehog.This paper focuses on exploring the expression and clinical significance of DNAH17 in laryngeal squamous cell carcinoma,aiming to provide a reference for the early diagnosis and personalized treatment of LSCC.Purpose To explore the expression of DNAH17 in LSCC and to analyze its relationship with the clinicopathological features of the patients.Method(1)Differentially expressed genes were screened in head and neck squamous cell carcinoma(HSNC)and normal head and neck tissues from the Cancer Genome Atlas(TCGA)and analyzed the expression of DNAH17 mRNA and protein in HSNC and normal head and neck tissues using UALCAN database;(2)Paraffin samples were collected from 68 cases of LSCC and 44 cases of normal laryngeal tissue adjacent to cancer.The expression of DNAH17 in paraffin samples was detected by immunohistochemistry,and its relationship with the clinicopathological features of LSCC was analyzed.Result(1)The high expression of DNAH17 in LSCC was 47.1%,which was significantly higher than in adjacent tissue(27.3%,P<0.05);(2)The expression of DNAH17 protein was significantly correlated with clinical stage of LSCC and tumor lymph node metastasis(all P<0.05),which was not associated with LSCC patients’ age,sex,site of onset,histological grade and depth of invasion(all P>0.05).Conclusion The expression level of DNAH17 protein was significantly increased in LSCC,which could be used as a potential molecular biomarker for the pathological diagnosis of LSCC.
