| Objectives: To detect the expression of Bcl-2,E-cadherin,N-cadherin proteins inlaryngeal squamous cell carcinoma,and to detecte the relationships among the threeproteins of LSCC clinical pathological, and then to find some tumor markers whichmight local invasion and lymphatic metastasis of LSCC to be used in clinical. Explorenew methods of diagnosis and prevention to provide a theoretical clinical reference.Meterial&Methods: The tissue of laryngeal squamous cell carcinoma samplewere obtained from47patients who underwent surgery pathological paraffin blocks inthe Tumor Hospital of Hunan Province as for experimental group, meanwhile26patients vocal polyp specimens from Department of Pathology University of SouthChina,autopsy specimens40cases normal laryngeal mucosa from judicialidentification center of University of South China as for the control group. Every fivewhite slice paraffin specimens, for HE, immunohistochemistry, and negative control.Bcl-2,E-cadherin,N-cadherin proteins were detected by S-P immunohistrochemistrystaining in between laryngeal squamous cell carcinoma and non-squamous cellcarcinoma of the larynx (the atypical hyperplasia tissue adjacent to carcinoma, vocalcord polyps and normal laryngeal squamous epithelium), with PBS take the place ofprimary antibody as a negative control.Results:1. The positive rates for Bcl-2protein of the Laryngeal squamous cell carcinoma,adjacent atypical hyperplasia, vocal cord polyps and normal laryngeal mucosawere respectively61.7%(29/47)ã€33.3%(11/33)ã€19.2%(5/26)ã€0(0/40). Thepositive rate for Bcl-2protein of the LSCC were higher than that of the adjacentatypical hyperplasia, vocal cord polyps as well as the normal laryngeal mucosa(P<0.05),and this difference is significant. The expression of Bcl-2protein was correlated with nvasion depth, degree of differentiation,clinical stage and lymphnode metastasis(P <0.05), and unrelated with age,sex of LSCC(P <0.05).2. The positive rates for E-cadherin protein of the Laryngeal squamous cellcarcinoma, adjacent atypical hyperplasia, vocal cord polyps and normal laryngealmucosa were respectively40.4%(19/47)ã€66.7%(22/33)ã€84.7%(22/26)ã€100%(40/40). The positive rate for E-cadherin protein of the LSCC were lowerthan that of the adjacent atypical hyperplasia, vocal cord polyps as well as thenormal laryngeal mucosa(P <0.05),and this difference is significant(P <0.05).The expression of E-cadherin protein was related to tumor invasion depth, degreeof differentiation, clinical stage and lymph node metastasis (P <0.05), andunrelated with age,sex of LSCC (P>0.05).3. The positive rates for N-cadherin protein of the Laryngeal squamous cellcarcinoma, adjacent atypical hyperplasia, vocal cord polyps and normal laryngealmucosa were respectively70.2%(33/47)ã€42.4%(14/33)ã€11.5%(3/26)ã€0(0/40). The positive rate for N-cadherin protein of the Laryngeal squamous cellcarcinoma were higher than that of the adjacent atypical hyperplasia, vocal cordpolyps as well as the normal laryngeal mucosa(P <0.05),and this difference issignificant (P <0.05). The expression of N-cadherin protein was related to tumorinvasion depth, degree of differentiation, clinical stage and lymph nodemetastasis(P <0.05), and unrelated with age,sex of LSCC (P>0.05).4. The expression rates of Bcl-2,E-cadherin,N-cadherin proteins in laryngealsquamous cell carcinoma.The expression of Bcl-2protein were negativelycorrelated with the expression of E-cadherin protein(r=-0.421,P=0.003), butpositively correlated with the expression of N-cadherin protein(r=0.635, P=0.000). But there was a negative effect correlation between positive proteinexpression of E-cad and N-cad in LSCC (r=-0.411, P=0.004).5. Survival analysis wan performed for47cases with the survival time.The patientswith Bcl-2protein strongly positive expression has shorter survival time thannegatives, negative expression of E-cadherin protein patiens and N-cadherinprotein positive expression were significantly increased mortality. Conclusion:1. The higher expression of Bcl-2,N-cadherin protein and the lower expressionof E-cadherin in the LSCC.2. Bcl-2, E-cadherin and N-cadherin proteins of expression are related withclinical stage of LSCC,malignant tumors,the extent of lymph node metastasisand Survival.3. United detection of the expression of Bcl-2, E-cadherin and N-cadherinproteins is used to predict metastasis and to judge prognosis of LSCC. |
