| Objective: Obstructive sleep apnea syndrome(OSAS)is a sleep-related breathing disorder which mechanism is closely related to upper airway stenosis.Fat accumulation in the pharynx is one of the important factors leading to upper airway stenosis in OSAS patients.Adipose tissue has endocrine function and adipose tissue in different parts has different functions.In this study,the experiment of proteomics was conducted on pharyngeal fatigue tissue in patients with severe OSAS,neck subcutaneous fat in patients with severe OSAS,and pharyngeal fatigue tissue in patients with mild OSAS to explore the difference and significance in protein expression between pharyngeal fatigue tissue and subcutaneous fatigue tissue in patients with OSAS.Methods: The adipose tissue samples of the subjects were divided into 3 groups.Group A: pharyngeal adipose tissue of patients with severe OSAS;Group B: subcutaneous fat in the neck of patients with severe OSAS;Group C: pharyngeal adipose tissue of patients with mild OSAS;Label Free proteomics technology was used to detect the proteome of these adipose tissue.The differentially expressed proteins between group A and group B were compared and bioinformatics analysis including significant difference analysis,cluster analysis,GO annotation,KEGG annotation,GO and KEGG enrichment analysis,protein interaction network and other bioinformatics analysis were performed.The same methods were used for differentially expressed proteins between group A and group C.Results: A total of 1232 differentially expressed proteins were identified in this experiment(FC>1.5,or<0.7,P<0.05).There were 1193 differentially expressed proteins including 864 up-regulated proteins and329 down-regulated proteins were identified between group A and group B,among which the expressions of CTSB and GFAT1 which are reverent to glycolipid metabolism were significantly up-regulated ingroup A,but the expressions of PLIN-1,LIPE and CD36 which are related to glycolipid metabolism were significantly down-regulated ingroup A;Besides,There were 39 differentially expressed proteins including 21up-regulated proteins and18 down-regulated proteins were identified between group A and group C,among which the expression of FBP2 which is a insulin resistance-related protein was significantly up-regulated ingroup A,The expression of WDR13 which is a insulin resistance-related protein was significantly down-regulated in group A.Conclusion: 1.Expressions of Proteins of pharyngeal fatigue tissue are different from subcutaneous fatigue tissue in patients with OSAS;2.The changes of expression of these differential proteins may be related to the occurrence of metabolic related complications of OSAS. |
