| Objective:To analyze the expression of ADHEF1 in stomach cancer and its clinical significance by combining database and basic experiments.Methods : Surgical specimens of gastric cancer patients who underwent surgical treatment at the Affiliated Hospital of Inner Mongolia Medical University between October 2020 and October2021 were collected,including 32 pairs of gastric cancer tissues and paired paracancerous tissues.Immunohistochemical staining was used to detect the expression of ADHFE1 protein in gastric cancer and to analyze the relationship between ADHFE1 protein expression and clinicopathological features.Fifteen pairs of frozen tissue samples were selected for q RT-PCR experiments to analyze the m RNA expression of ADHFE1 in gastric cancer;the expression datasets of ADHFE1 in GEPIA and Oncomine databases were mined to analyze the difference of its expression between different tumors;and to analyze whether there was differential expression in gastric cancer;the methylation data were collected from TCGA database to analyze the difference between ADHFE1 levels in stomach tumors and normal tissues;String database analyze ADHFE1-interacting protein,linkedomics database analyze ADHFE1-interacting gene and do GO enrichment and KEGG pathway analysis.Results:Analysis of the staining results showed that ADHFE1 was mainly expressed in the cytoplasm,and most of them appeared brownish yellow.The positive rate of ADHFE1 protein in 32 paraneoplastic tissues was 59.4%,which exceeded the positive rate of31.3% in 32 gastric cancer tissues.the expression of ADHFE1 protein correlated with the depth of tumor infiltration(P=0.026<0.05)and lymph node metastasis(P=0.028<0.05).q RT-PCR showed that the expression of ADHFE1 m RNA in gastric cancer tissues was 1.024±0.226 and 2.251±0.450 in paired paraneoplastic tissues,with statistical differences in expression(P<0.05).Multiple databases showed that ADHFE1 expression levels were lower in gastric cancer than in normal tissues;TCGA database download analysis obtained elevated methylation expression of ADHFE1 compared to normal tissues;Enrichment analysis showed that ADHFE1 was mainly present in the cytoplasm and had catalytic activity,The enrichment analysis showed that ADHFE1 is mainly found in the cytoplasm and has molecular functions such as catalytic activity,cytokine binding,participation in the signaling pathway of phospholipase C-activated G protein-coupled receptors,mitochondrial gene expression and other processes as well as regulation of various biological processes including cell cycle.Conclusion:ADHFE1expression was decreased and methylation level was increased in gastric cancer,and the expression level of ADHFE1 correlated with the degree of gastric cancer infiltration and lymph node metastasis.ADHFE1 may be used as a new biomarker in the diagnosis and treatment of gastric cancer. |
