ObjectiveSerological indexes,clinical characteristics and maternal and infant outcomes of severe preeclampsia were analyzed.Detection of NLRP3 inflammatory bodies and apoptosis-associated speck-like protein containing in placental tissues of severe preeclampsia patients and normal pregnant women The expression of ASC,caspase-1,interleukin-1β(IL-1β)was analyzed to investigate their correlation with clinical serological indexes and their possible role in the pathogenesis and development of severe preeclampsia.MethodFrom October 2019 to June 2021,a total of 95 pregnant women with single-child pregnancies who were hospitalized in the obstetrics department of the Affiliated Hospital of Qingdao University and had elective caesarean sections were collected,including 30 pregnant women with severe PE(research group)and 65 pregnant women with normal blood pressure(control group).The clinical data and serological related indicators were recorded,and the m RNA expression levels of NLRP3 inflammasomes,ASC,Caspase-1 and IL-1β in the two groups of placental tissues were compared with Western blot and RT-q PCR,and the correlation between NLRP3 genes and protein expression levels and clinical serological indicators was analyzed.SPSS21.0 statistical software was used for data analysis,and the independent sample t-test,χ~2 test,rank and order test were used for statistical analysis,and pearson correlation coefficient was used to test the correlation between quantitative variables.Results1.1.The expression levels of NLRP3 inflammasome,ASC,Caspase-1 and IL-1βin placenta in severe preeclampsia group were significantly higher than those in normal pregnancy group(P<0.05).The m RNA expression levels of NLRP3 inflammasome,caspase-1 and IL-1β in placenta were significantly higher than those in normal pregnancy group(P<0.05).2.Platelets and Platelet/Lymphocyte ratio(PLR)in severe preeclampsia group were lower than those in normal group.Neutrophil Lymphocyte ratio(NLR)of Neutrophil and Neutrophil/Lymphocyte ratio(NLR)were higher than those of normal pregnancy group,the differences were statistically significant(P<0.05).3.Serum uric acid level in severe preeclampsia group was significantly higher than that in normal group,and there was a positive correlation between serum uric acid level in peripheral blood and NLRP3 protein expression level in placental tissue(r=0.88,P<0.05).There was a positive correlation between NLRP3 m RNA expression and placental tissue(r=0.92,P<0.05).4.Serum cholesterol level in severe preeclampsia group was significantly higher than that in normal group,and there was a positive correlation between cholesterol level and NLRP3 protein expression level in placental tissue(r=0.84,P< 0.05).There was a positive correlation between cholesterol and NLRP3 m RNA expression in placental tissues(r=0.87,P<0.05).5.There was no significant correlation between NLRP3 inflammatory body protein and m RNA expression levels and neutrophil count,platelet,NLR and PLR in severe preeclampsia group(P>0.05).ConclusionNLRP3 inflammasome activation may be involved in the pathogenesis and development of severe preeclampsia,and high uric acid and cholesterol levels in patients may be one of the important reasons for NLRP3 inflammasome activation.
