This study investigated the species of Ixodes in sheep in Shawan City,Xinjiang,and analyzed the bacterial flora structure of Rhipicephalus sanguineus and its infections with the Anaplasma ovis,Candidatus Rickettsia barbariae,and Coxiella burnetii.The results are as follows.⑴ The results showed that the common ticks on the surface of the local sheep were Rhipicephalus sanguineus,Rhipicephalus turanicus,and Haemaphysalis punctate,they were determined by morphological characteristics and molecular biology methods.⑵ The results of high-throughput showed that there were 11 phyla and 140 genera of bacteria in Rhipicephalus sanguineus,among which Proteobacteria,Firmicutes,and Bacteroidota were the most abundant phyla.The highest relative abundance genera were Acinetobacter,Delftia,and Stenotrophomonas.The relative abundance of Anaplasma,Rickettsia,and Coxiella was 0.095%,2.926%,and 0.02%,respectively.The positive rates of Anaplasma ovis,Candidatus Rickettsia barbariae,and Coxiella burnetii were 44.7%,77.6%,and 85.9%,respectively.⑶ The glt A,gro EL,and icd gene sequences were amplified to analyzed phylogenetic relationship,and concluded that Rhipicephalus sanguineus carried Anaplasma ovis,Candidatus Rickettsia barbariae,and Coxiella burnetii.Quantitative PCR results were as follows,the infection intensity of each positive Rhipicephalus sanguineus infected with Anaplasma ovis,Candidatus Rickettsia barbariae,and Coxiella burnetii was 4003.2±2109.6 copies,104491.2±75649.2 copies,267.6±235.2 copies,respectively.
