To study the effects of heavy metals to chicken caecum microbial diversity.72 healthy 30-day-old Hy-line layer were selected,and randomly divited into 4 groups:control group,CuCl2(150mg/L)group,HgCl2(140mg/L)group,PbCl2(350 mg/L)group.Each group with 3 repeats,and each repeat with 6 layers.Cecum and cecal contents were collected on days 30 th,60th,and 90 th.Observe the chicken caecum structure and using Illumina-Hiseq 250 high-throughput sequencing techonlogies to sequence the microbial community structure and diversity.the results showed that:1.The histology of the cecum in the CK group was normal,which showed complete morphological structure,mucosal integrity,distribution,and glands.Compared with the control group,Cu group,Hg group had a small amount of lymphocyte hyperplasia.Pb group of intestinal mucosa has a large number of shedding,and the lamina propria has a large number of lymphocyte proliferation.2.72 chickens gut microbe standardized to 5527565 sequences,2431 OTUs,representing 28 phyla,348 genera.3.The most microbe was classified five Bacterial phyla in chicken distal gut flora,respectively:Firmicultes,Bacteroides,Verrucomicrobia,Proteobacteria,Teneriactes.Bact-eroi des were the largest proportion at the genus level,accounted for the total bacterial genus 19.2%.4.Firmicultes was the most abundance phylum at the phylum level,lactobacillus and coprococcus was the the most abundance genus at the genus level in the Cu group.5.Teneriactes was the most abundance phylum at the phylum level,weissella and erysipelotrichaceae was the the most abundance genus at the genus level in the Hg group.6.The abundance of Firmicultes and Bacteroides decreased,prevotella,weissella and erysipelotrichaceae were the the most abundance genus at the genus level in the Pb group.In conclusion,our study showed that heavy metals had a certain damage to the cecal tissue of chickens and affected the structure and diversity of intestinal flora.
