| The appearance of the guinea pig is cute,smart,lively and docile,and it has become a pet that people all over the world compete to raise.When guinea pigs suffer from secretory otitis media,they will show poor response to surrounding sounds,which will affect the interaction with people.At the same time,it will easily irritate and scratch their ears,which will seriously affect the health of people and guinea pigs.Quercetin has a wide range of pharmacological effects.At present,there is no report on whether quercetin can prevent secretory otitis media in guinea pigs.Therefore,in this experiment,inactivated Streptococcus pneumoniae was used to establish a model of secretory otitis media in guinea pigs,and whether quercetin has a preventive effect on secretory otitis media in guinea pigs was studied,so as to provide experimental basis for the prevention of secretory otitis media in guinea pigs.24 healthy male guinea pigs were randomly divided into control group,model group and quercetin group,with 8 pigs in each group.The control group and model group were given0.1ml/10 g body weight of normal saline by gavage,while the quercetin group was given0.1ml/10 g body weight of 2% quercetin suspension by gavage once a day for 3 consecutive days.On the fourth day of the experiment,inactivated Streptococcus pneumoniae suspension was injected into the right tympanum of each guinea pig through the anterior and inferior tympanic membrane to establish a model of secretory otitis media,and the control group was injected with the same amount of normal saline into the right tympanum by the same method,and then continued to give it for 4 days.On the 7th day after modeling,tympanic membrane photography,ABR audiometry,acoustic immittance detection,and middle ear effusion were taken to detect inflammatory factors.Then the guinea pigs were killed by decapitation,the right auditory vesicle was taken,fixed with 4% paraformaldehyde,decalcified with 0.5M EDTA,embedded in OCT,and observed under conventional HE staining light microscope.The results of this experiment show that compared with the model group,the tympanic membrane of guinea pigs in quercetin group is slightly turbid,with less or no congestion,and there is a little or no effusion in the tympanic cavity;ABR response threshold decreased,and I wave latency and III wave latency shortened,with statistical significance(P<0.01).The tympanogram produced by acoustic immittance test showed that the tympanogram in the model group was flat,and the peak amplitude was basically 0mmho.The tympanogram in the quercetin group had a peak,and the average tympanogram pressure was(-37.00±41.47)da Pa,and the peak amplitude was(0.07±0.03)mmho.The differences in tympanogram pressure and peak amplitude were statistically significant(P<0.05).The contents of inflammatory factors IL-1β,IL-6 and TNF-α all decreased,and the differences were statistically significant(P<0.05).The middle ear mucosa is thinner and there are fewer neutrophils,and the difference is statistically significant(P<0.05).Compared with the control group,the tympanic membrane of guinea pigs in quercetin group has not reached the normal state;ABR threshold was increased,and the difference was statistically significant(P<0.01).The latency of I wave was prolonged with statistical significance(P<0.01).The tympanogram of quercetin group has a peak,the average tympanic pressure is(-37.00±41.47)da Pa,and the peak amplitude is(0.07±0.03)mmho.Compared with the control group,the difference of tympanic pressure is statistically significant(P<0.05),but the difference of peak amplitude is not statistically significant(P>0.05).The contents of inflammatory factors IL-1β and IL-6 were higher,the difference was statistically significant(P<0.01),and the content of TNF-α was also higher,the difference was statistically significant(P<0.05).The middle ear mucosa is thicker and there are more neutrophils,and the difference is statistically significant(P<0.01).In this study,quercetin can obviously reduce the hearing loss caused by secretory otitis media in guinea pigs,reduce the thickness of middle ear mucosa and neutrophil infiltration,and inhibit the expression of IL-1β,IL-6 and TNF-α in middle ear effusion,but it is not up to normal level. |
