This research aimed to study the epidemic of Salmonellosis in both layer and breeding chickens on different age selected from Shanxi province. The samples were analyzed by normal biochemistry and PCR test, and the attenuated vaccine was developed according to the test results to prevent the Salmonellosis in chickens in Shanxi province.The related samples were collected from more than 10 different large farms. More than 100 samples were separated from the chickens, primes of variable region of flic gene of Salmonella pullorum and Salmonella gallinarum were designed to Select 19 samples randomly from the above. The results showed the PCR product of 12 trains were more or less than 600bp, the positive rate of Salmonella pullorum and Salmonella gallinarum is 63%. 2 of these 12samples were random selected and were co-cultured with two standard strains- C79-13 and C70-20, and inactivated by methanal to aluminium hydroxide adjuvant vaccine, which was then used to immune healthy chickens. The antibody level reached the highest level after 15days, and the T lymphocyte and E chaplet detection also showed that the cellular immune level was obviously higher in immunity group than that of control group. The survival rate was 90.5%under 10LD dose of virulent strain challenge and the separation rate of Salmonella in immune group was 2.1%, which all showed the vaccine had satisfactory security and immune effect.
