| The purpose of this study was to explore the association of FSHR and ESR gene polymorphisms with egg laying performance,egg quality and hatching performance of Baicheng oil chickens,and to screen the SNP loci related to the reproductive traits of Baicheng oil chickens,in order to select and breed from the molecular level.Baicheng oil chickens fecundity strains provide scientific basis.In the experiment,150 healthy Baicheng oil chickens hens were randomly selected as research objects,and their egg production performance,egg quality and hatching performance were recorded and measured,and DNA was extracted from blood for FSHR,ESR gene polymorphism detection.Correlation analysis between FSHR,ESR gene polymorphisms and reproductive performance of Baicheng oil chickens,The results showed that:(1)The results of the correlation analysis of the reproductive performance of Baicheng oil chickens showed that the total egg production of Baicheng oil chickens at 300 days old and the average egg production weight of 300 days old showed a very significant positive correlation(P< 0.01),the total egg production at 300 days old showed a significant negative correlation with the age of the opening day and the weight of the eggs(P< 0.01),and the total egg production of 300 days old was positively correlated with the starting weight,and the correlation was significant(P< 0.05)(2)The results of genetic diversity analysis of SNPs showed that g.7640639 T >C,g.7692270 C >T,g.7698478 A >G,g.7716725 G >C,g.7716470 T >C,g.48747211 T >G,g.48777663 C >T,g.52521976 T >C and g.52541181 C >T loci polymorphism information content showed moderate polymorphism(0.25< PIC<0.50),indicating that the above SNPs all have high genetic potential.(3)The correlation analysis between the SNPs of FSHR gene and the reproductive performance of Baicheng oil chickens showed that the date of birth of CC type was significantly higher than that of TT type at SNP2 locus(P< 0.01),significantly higher than that of CT type(P< 0.05);the egg weight of TT type was significantly higher than that of CT type(P< 0.05),and the dead embryo rate of incubated eggs was significantly higher than that of CC type(P< 0.05).The thickness of eggshells of TT type was significantly higher than that of CC type at SNP3 locus(P< 0.05).The total egg production at 300 days of age of CG type was significantly higher than that of CC and GG types at SNP5 locus(P< 0.01),the average egg weight and egg laying weight at 300 days of age were significantly higher than that of GG type(P< 0.05),the birth weight was significantly higher than that of CC genotype(P< 0.05),and the thickness of eggshells of GG genotype was significantly lower than that of CC and CG types(P< 0.05).(4)The results of the correlation analysis between the SNPs of ESR gene and the reproductive performance of Baicheng oil chickens showed that the SNP loci at exon2 and exon4 of ESR1 gene had extremely significant effects on the total egg production at 300 days of age(P< 0.01).Egg production had a significant effect(P< 0.05),and the total egg production of individuals with GG genotype at exon2 and TT genotype at exon4 at 300 days was significantly higher than those of GT and CT types.The SNP loci of ESR2 gene exon1,exon2 and exon4 had a significant effect on the hatching rate(P< 0.05).The SNP loci at exon1 and exon2 had a very significant effect on the hatchability of fertilized eggs(P< 0.01),and the SNP loci at exon4 and exon8 had a significant effect on the hatchability of fertilized eggs(P<0.05).The homozygous genotypes TT and CC fertilized eggs have the highest hatchability.The SNP loci at exon1 and exon2 had a significant effect on the dead embryo rate of hatched eggs(P< 0.05).In summary,through the correlation analysis between FSHR,ESR gene and Baicheng oil chickens reproductive performance,it can be found that the SNPs loci of FSHR and ESR genes have a significant impact on the reproductive performance of Baicheng oil chickens,and then the marker gene loci that may affect the reproductive performance of Baicheng oil chickens are screened,which provides a theoretical basis for exploring the molecular genetic mechanism affecting the reproductive traits of Baicheng oil chickens. |
