Impatiens hybrid cv.sunpatience is a new hybrid of Impatiens,which belongs to Impatiens hyfamily and Impatiens genus.It has beautiful flower shape and long ornamental period.The production of Impatiens balsamina in sunpatience mainly relies on the import of rootless seedlings.Repeated cutting causes serious variety degradation and affects the potted quality.In this study,explants selection,callus induction,somatic embryogenesis and rooting in vitro were carried out to optimize the tissue culture system of Impatiens sunpatience:1.Establishment of tissue culture and rapid propagation system of Impatiens sunpatience:With 1~2 cm size of lateral buds,sterilization 9 min,stripping into 0.3~0.4 mm stem tip survival rate is the highest.the optimal medium for shoot tip growth was MS+0.5mg/L 6-B A+0.1 mg/L NAA.2.Effects of explant types and culture methods on callus induction and embryogenic callus induction:(1)The optimal callus induction medium was:Leaf blade as material:MS+0.5mg/L 6-B A+1.0mg/L 2,4-D+0.2mg/L NAA+light culture.Stem segment as material:MS+0.5mg/L 6-B A+1.0mg/L 2,4-D+0.2mg/L NAA+light culture.Root tip material:MS+0.5mg/L 6-B A+1.0mg/L 2,4-D+0.2mg/L NAA+dark cultured.When cluster bud induction was conducted on callus,:MS+1.5mg/L TDZ+0.2mg/L NAA+dark cultured.(2)The best induction medium of embryogenic callus were respectively:Leaf blade as material:MS+1.5mg/L PIC+0.2mg/L NAA+light culture.Stem segment as material:MS+2.5 mg/L PIC+0.2mg/L NAA+light culture.Root tip material:MS+1.5mg/L PIC+0.2mg/L NAA+light culture.Use of embryogenic callus for somatic embryogenesis:MS+1.5mg/L PIC+0.2 mg/L NAA+light culture.Use of Light Culture in Somatic Embryo GerminationMS+0.5mg/L 6-BAThe highest germination rate.3.Techniques of rooting ex vitro of Impatiens sunpatience:The best rooting effect was obtained when the ratio of peat to vermiculite was 3:1.The rooting rate of 1~4 generations of tissue culture seedlings was better than that of cultivated seedlings.With the increase of heading generations,all aspects of indicators were decreased,the first generation traits were the best,the fourth generation traits were the worst,but the fourth generation seedlings were better than cutting seedlings.
