Anoectochilus roxburghii(Wall.)Lindl,a genus of perennial herb,is a perennial herb with high value in medicinal and ornamental aspects,but it is severely damaged now,forcing wild resources to be scarce.The use of tissue culture technology has important research significance and economic value for the preservation and expansion of its germplasm resources.In this experiment,we studied the effects of different concentrations and different kinds of plant growth regulators on their proliferation,the effects of different media on their rooting,and the effects of different substrates on the survival rate of transplanting.Base and growth matrix.The sterile seedlings of A.chinensis were used as experimental materials,and MS was used as the basic medium.The experimental results are as follows:1.Primary cultureThe optimum medium for the primary culture of A.chinensis was MS+6-BA4.0mg/L+NAA0.1mg/L,the average plant height was 3.0cm,the differentiation rate was 100%,and the multiplication ratio was 6.676.2.SubcultureThe most suitable medium for subculture of A.chinensis was MS+CPPU 5.5 mg/L+NAA 0.1 mg/L,the average plant height was 3.7 cm,the differentiation rate was 100%,and the multiplication ratio was 7.5.3.Rooting cultureThe most suitable medium for rooting of A.chinensis was 1/2MS+NAA0.5 mg/L+IBA 1.0 mg/L+2g/L activated carbon,the average plant height was 4.28 cm,and the rooting rate reached 100%.4.TransplantingDuring the transplanting process,the best and fastest substrate for the growth of A.acuminata is vermiculite,with a survival rate of 95%.
