The Study Of The Function Of Ver2 Gene In Wheat Albino Line

The albinism of Wheat albino line was induced by low temperature and regulated by nuclear genes and cytoplasmic genes.Gene chips and digital gene expression profiling of Wheat albino line and Aibian NO.1 shown that the expression of Ver2 gene was distinctly different in the two strains.In this study,we put emphasis on the expression characteristics of Ver2 and molecular characteristics analysis of its promoter.We also studied the relationship between Ver2 gene and the albinism of Wheat albino line.The main results are as follows:1.Ver2 gene existed in many common wheat varieties and widespreading.These varieties had no winter and spring biological specificity.2.The VER2 protein in Wheat albino line and other common wheat varieties was expressing at room temperature and it had tissue specificity.It largely expressed in interior leaves.3.By cloning and sequencing,we found that Ver2 promoter had intact small and larger repeat.In addition,we had cloned a mutational sequence,in which there was a deletion from-286 to-1897 and included many important elements.4.We counted the phenotypes and Ver2 gene of F2(F×CS)and F2(F×A)groups.From the result we found that both albinistic and green plants had Ver2 or not.It indicated that Ver2 is not decisive for albinism;however,we found that 5 significantly albinism plants all had Ver2 gene,so we speculated that Ver2 may promote wheat albinism.5.We constructed the Ver2 gene plant expression vector and successfully transformed in arabidopsis and tobacco.Lastly we had got stable transgenic plants.In conclusion,the origin of Ver2 in Wheat albino line was complicated.Ver2 didn’t directly affect the albinism of Wheat albino line,but it may promote the albinism.The structure mutation of Ver2 promoter may change its expression pattern.
