Preliminary experimental results suggests that epigenetic regulates wheat albescent system of whitening process.We analyze the tesults of gene microarray,The expression pattern of epigenetic genes are analyzed by Real time PCR;TSA and 5-aza treat the albinism line of wheat in different conditions to observe the phenotypic changes;The histone acetylation level is detected by western bloting,albinism albino system further determines whether they are associated with DNA methylation and histone acetylation.Following conclusions:DNA methylation are involved in whitening process of albescent lines.Under low temperature,DNA methyltransferase gene TaDRM2 of albinism line increase expression and they are more than Aibian1,but TaMET1?TaCMT are downregulation.Then the exprission of DNA methyltransferase gene of green seeding of albinism line is similar to Aibian1.50?mol/L 5-azaC promots albino in albinism line.Histone acetylation are involved in whitening process of albescent lines.In the albino stage,Albinism system histone deacetylase gene as HDAC2,HDAC4,HDAC3 and HDAC6 increase expression and they are more than Aibian1;But the expression pattern of histone acetyltransferase genes have the same situation.Then the exprission of histone modification enzyme of green seeding of albinism line is similar to Aibian1.albinism line and Aibian1 are treated with TSA(400nmol/L)and low temperature,which inhibite albino in albinism line,however,the leaf color of Aibian1 no have change.By Western Blot detecting the histone acetylation levels,under low temperature,histone acetylation levels of albinism line is lower than Aibian1,which evidence the preliminary results.Increasing the histone acetylation level of albinism line delay albino.The different histone acetylation levels regulates expression of gene,which case the same genotype albinism line having different phenotypes.Sin3 is a co-repressor of transcription factor binding with HDAC,however,Sin3 of the wheat don't have relevant reports.Through literature and sequence alignment,we speculate that Sin3 is combined with HDAC3 in wheat.Real-time PCR determine similar expression patterns of Sin3 and HDAC3 in wheat.So we sumise that Sin3 and HDAC3 interact to form Sin3 / HDAC complex,which regulates expression of downstream genes.
