Tree peony is gorgeous in color and diverse in form,which has high ornamental value.As a new type of oil crop,peony has been developed unprecedentarily.Paeonia ostii ‘Feng Dan’ is the main cultivar of oil tree peony.During production,a large number of peony seed coat and other by-products have not been used effectively.It is important to make full use of seed coat appendages to improve the comprehensive benefit of tree peony.In this paper,the main active substances of Paeonia ostii ‘Feng Dan’ seed coat were extracted and evaluated,hoping to provide a theoretical basis for the comprehensive development and utilization of the by-products of oil tree peony.The main results achieved are as follows:1.HPD-100 macroporous resin column chromatography was used to extract the seed coat of Paeonia ostii ‘Feng Dan’ by gradient elution,and nine components(Z-2 ~ Z-10)were enriched.The results of activity experiments showed that all the elution layers had strong antioxidant and anti-neuritis ability.The antioxidant capacity of Z-2 to Z-5 elution layers were better than other elution layers,and the inhibition rate of NO production by microglia in BV-2 mice induced by LPS was higher in Z-2,Z-3 and Z-5 elution layers,which could reach more than 75%.2.A total of 76 compounds were identified by non-targeted metabolomics analysis of different eluent fractions of Paeonia ostii ‘Feng Dan’ seed coat extracts,mainly stilbenes,phenols and flavonoids.Analysis of non-targeted metabolomics quantitative data showed that stilbenes are the major bioactive components in the seed coat.3.Five stilbenes,suffruficosol A,suffruficosol B,suffruficosol C,trans-ε-viniferin and trans-gnetin H,were isolated and purified from the macroporous resin elution layer fractions.The total amount of stilbenes in different elution layers of macroporous resin was determined.The total amount of stilbenes increased first and then decreased,with a peak of88.85 g/100 g in Z-3 eluting layer.4.Single factor test,Box-Behnken test and response surface analysis were performed to optimize the extraction process of stilbenes from Paeonia ostii ‘Feng Dan’ seed coat.The optimal extraction process of stilbene was obtained: time 40 min,ethanol concentration was70% and solid-liquid ratio was 1:20,extraction content of stilbene was 121.26 mg/g and the extraction yield was 81.54%.
