| In the present study,the peony seed oil was extracted by using supercritical CO2 fluid.The response surface optimization method was used to determine the optimum extraction conditions of peony seed oil using supercritical CO 2 extraction.The optimum extraction conditions used were as follow;extraction time of 124 min,extraction temperature 46℃,extraction pressure 24 MPa,CO2 flow 21 L/h and 27.36% extraction rate.Beside s,the optimum process conditions of peony seed oil microcapsulation by spray drying were such as: oil/beta cyclodextrin ratio is of 1:8,emulsifier content 0.25% and the mixing time 2 h and the embedding rate of 69.86%.The peony seed oils were extracted by supercritical CO2 fluid,soxhet and mechanical screwpress method,respectively.Obtained peony seed oil mainly consisted of palmitic,stearic,oleic,linoleic and linolenic acid detected by GC-MS analysis,and the content of unsaturated fatty acid were observed to be more than 89%.The extraction methods had few effect on the fatty acid composition in peony seed oil.The mechanical screwpress oil possessed a strong radical scavenging ability,whereas,soxhet oil had remarkable alpha-amylase,alpha-glucosidase inhibition and highest activation energy.The protein content of peony seed meal reached up to 31.40%,which mainly consisted low molecular weight protein subunits.The proportion of essential amino acids in total amino acid content was 35.77 mg/g.T he water-binding capacity was observed to be better than oil binding capacity at room temperature.The lowest solubility,emulsification and foaming ability observed were at p H 5.0.Two phase transition of protein isolated occurred in temperature range of 30-160℃.Hydrolysis dynamics model of Peony seed protein was DH=3.55ln[1+(2.56E0/S0+0.12)t].Subsequently,880.25(M+H+)antioxidative peptide sequence for SMRKPPG was isolated and further purified by using enzymolysis,ultrafiltration,anion chromatograph y,gel chromatography and RP-HPLC. |
