At present,the main preparation methods of peony seed oil by squeezing method,but in this way will produce a lot of peony seed shell and peony seed cake.Research and development of these by-productscan can further improve the added value of peony seed.This article first part preliminary explored chemical composition of peony seed shell and peony seed cake of Paeonia ostii.We try to use way of Sephadex LH-20,preparative HPLC,silica column chromatographies,Positive phase silica gelcolumn and thin Layer Chromatography for the effective parts of extracts of separation.Using spectral data analysis to identify the structure.We finally got 18 times compounds from peony seed cake of Paeonia ostii.They are elucidated as follws: 2,3-dihydroxymudanpioside G(1),10-hydroxy-ziziphoroside C(2),Mudanpioside F(3),10-hydroxypaeoniflorin(4),Isomaltopaeoniflorin(5),4’’-hydroxy-4-methoxy-paeoniflorin(6),4’’-hydroxy-paeonidanin(7),Albiflorin R1(8),Pyridypaeoniflorin(9),1-D-glucopyransoylpaeonisuffrone(10),8-debenzoylalbiflorin(11),8-debenzoylpaeoniflorin(12),4,9-dihydroxy-8,10-dehydrothymol-1-O-β-Dglucosides(13),(8R)-Pigeritone-4-en-9-O-bdglucopyranoside(14),Paeonidanin(15),Albiflorin(16),paeoniflorin(17),Oxypaeoniflorin(18)and Compound 1,Compound 2,Compound 4 were identified as a new compounds.From peony seed shell of Paeonia,We finally got 8 times Stilbenoids,as follws: suffruticosol A(Ⅰ),suffruticosol B(Ⅱ),suffruticosol C(Ⅲ),Transsuffruticosol D(Ⅳ),Gnetin H(Ⅴ),Cis-ε-Viniferin(Ⅵ),Trans-ε-viniferin(Ⅶ),AmpelopsinB(Ⅷ).In the latter half,to establish an HPLC method for simultaneous determination of six monoterpene glycosides in seed cake and seed shell of peony.We have found that peony seed cake monoterpene glycoside content is far greater than its content in the peony seed shell in the qualitative and quantitative analysis.We have found monoterpene glycoside compounds Isolated have anti-inflammatory activity of preliminary screening: Compound 4,Compound 5,Compound 6,Compound 7,Compound 8,Compound 9,Compound 16,Compound 17,Compound 18 have good antiinflammatory activity.Using the four different cancer cells to screen Isolated of stilbene Compound Ⅰ,Compound Ⅱ,Compound Ⅲ,Compound Ⅴ,Compound Ⅵ,Compound Ⅶ,finding all have certain inhibition,but the Compound Ⅱ have thesignificantly inhibition rate of A549 and 7721 as weel as the Compound Ⅴ have the inhibition rate of EC109 and the compound Ⅲ have the sinhibition rate of Hela.The study has been clear about the chemical composition of the oil with peony seed shell and the peony seed cake and the main active role,for further research monoterpene glycosides compounds and stilbene compounds provides a good research direction,and provide the scientific basis. |
